桃仁蛋白的单剂量及重复给药毒性试验

摘　　要：目的:通过桃仁蛋白单剂量及重复给药毒性试验,评价该药物用药安全性。方法:①单剂量毒性试验方法:在一日内单次给药。动物选择:昆明小鼠;给药途径:腹腔注射;②重复给药毒性试验方法:固定剂量每日连续给药。动物选择:W istar大鼠;给药途径:腹腔注射。结果:单剂量毒性试验结果提示,最大耐受剂量为300mg/(kg.d);重复给药毒性试验结果提示在给药期及恢复期对大鼠红细胞、白细胞、血小板等全血指标、谷草转氨酶、谷丙转氨酶、肌酐、尿素氮、总蛋白、白蛋白、胆固醇、血糖等血清指标均无明显影响;心、肝、脾、肺、肾等10个脏器大体标本外观、病理切片组织学结构无明显异常。结论:桃仁蛋白腹腔注射无急性和长期毒性作用。Objective：To evaluate safety pharmacology of peach seeds protein（psp） by single and repetitive dose toxicity testing.Methods：①To administrate mice a single dose of psp by peritoneal injection in a day in single dose toxicity testing;②To administrate Wistar rats quantitative and repetitive dose of psp in a month by peritoneal injection in repetitive dose toxicity testing.Results： It was concluded that the maximum tolerance dose of psp was 300mg/（kg·d） in single dose toxicity testing.It was also indicated that there was no significant effects of psp on whole blood WBC,RBC,Hb,serum AST,ALT,CREA,BUN,GLU,CHO,TP,ALB and the gross organs inspection and biopsy of heart,liver,spleen,lung,kidney in rats were normal in repetitive dose toxicity testing.Conclusion：There was no acute and chronic toxicity of psp in single and repetitive dose toxicity testing.
