摘  要:目的:建立脑微血管内皮细胞体外缺血再灌注损伤模型,研究细胞生存活性和NO含量的变化,探讨清开灵有效组分的保护作用。方法:体外培养大鼠脑微血管内皮细胞,氧糖剥夺法模拟缺血再灌注损伤,MTT法测定细胞生存活性变化并结合硝酸酶还原法测定细胞上清NO含量。结果:模型组细胞OD值明显低于正常组,NO含量明显升高(P<0.01);清开灵各组分都显著降低上清中NO含量,而且胆酸、黄芩苷、栀子苷(12.5μg.mL-1)组和新清开灵注射液(0.03%)明显增高OD值(P<0.05)。结论:清开灵有效组分通过提高细胞生存活性、减少NO分泌而保护体外缺血再灌注损伤的脑微血管内皮细胞。Objective: To establish an in vitro ischemia and reperfusion injury model of rat cerebral microvascular endothelial cells,observe the changes of cell survival activity and NO content,expecting to explore the mechanisms of QingKaiLing effective components protecting brain from injury of inflammatory cascade in cerebral ischemia diseases.Method: Primary culture of rat brain MVEC,establishing an in vitroischemia reperfusion model(OGD),applying MTT colorimetric method to observe the changes of cell survival activityand nitrate reductase method to study the effect of medicine on NO content of cultured cell supernate fluid.Result:The OD value of model group is lower than normal group,and NO content higher significantly(P〈0.01).To compare with model group,all components and new QingKaiLing injection can decrease NO contents(P〈0.05);In cell survival activity study(MTT),the OD value of geniposide,baicalin,cholic acid,hyocholic acid 12.5μg·mL-1and new QingKaiLing injection 0.03% group are significantly higher than model group(P〈0.05).Conclusion:The protective functionof QingKaiLing effective components on inflammatory cascade in cerebral ischemial diseases is probably by increasing cell survival activity and reducing NO contents.
