穴位敷贴对过敏性鼻炎小鼠血细胞分类和血清SIgE的影响

摘　　要：【目的】观察穴位敷贴对过敏性鼻炎模型小鼠外周血嗜酸性粒细胞(EOS)和血清卵白蛋白(OVA)特异性IgE(SIgE)的影响。【方法】选用BALB/C小鼠60只,随机分成5组:正常对照组、模型组、穴位敷贴组、激素阳性对照组(激素组)和磷酸盐缓冲液(PBS)阴性对照组(PBS组);除正常对照组以外,其他组均采用OVA致敏及攻击方法复制过敏性鼻炎模型,穴位敷贴组以中药(白芥子、玄胡、细辛、甘遂、苍耳子)敷贴大椎穴,激素组以100mg/L地塞米松腹腔注射,PBS组采用大椎穴假治疗同时腹腔注射PBS。以苏木素—伊红(HE)染色检测各组小鼠外周血EOS计数及百分率,酶联免疫吸附(ELISA)法检测各组小鼠血清SIgE水平。【结果】模型组小鼠外周血EOS计数与百分率、血清SIgE水平均显著性升高(P<0.05或P<0.01);穴位敷贴组和激素组的治疗均可显著性降低模型小鼠EOS计数与百分率及SIgE水平(P<0.05),两治疗组作用相仿(P>0.05)。【结论】穴位敷贴治疗过敏性鼻炎的作用可能与其能降低外周血EOS计数及血清SIgE水平有关。Objective To observe the effect of acupoints external application （AEA） of herbal drug on peripheral eosinophil （EOS） count and serum IgE level of the experimental murine model of allergic rhinitis induced by ovalbumin （OVA）. Methods Sixty BALB/C mice were randomized into 5 groups： normal group, model group, AEA group, hormone positive group and phosphate buffer solution （PBS） negative group. Murine models of allergic rhinitis were induced by sensitization and activation of OVA. AEA group received external application of powder of Semen Sinapis, Rhizoma Corydalis, Herba Asari, Radix Kansui and Fructus Xanthii on acupoint of Dazhui; hormone group received intraperitoneal injection of dexamethasone （ 100 mg/L ） ; PBS group received pseudo-acupoints external application and intraperitoneal injection of PBS. Hematoxylin-eosin staining was used to detect the EOS count and percentage in peripheral blood of different groups, and enzyme-linked immunosorbent assay （ELISA） was applied to detect the serum level of specific IgE （SIgE）. Results EOS count and percentage in peripheral blood and serum SIgE level were increased in the model group （P 〈0. 05 or P 〈0. 01 compared with the normal group） , and AEA and hormone treatment decreased the above indexes （ P 〈 0. 05 compared with the model group）. The effect in AEA group was similar to that in hormone group （ P 〉 0. 05 ）. Conclusion The therapeutic mechanism of AEA of herbal drug for OVA-induced allergic rhinitis may be related with the decrease of peripheral eosinophil count and serum IgE level.
