摘 要:【目的】探讨金钗石斛(Dendrobium nobile Lindl.)总生物碱和粗多糖对白内障的抑制作用。【方法】采用溶剂法分离提取金钗石斛总生物碱和粗多糖;体外培养大鼠晶状体,加入H2O2与总生物碱(或粗多糖)共培养24h;分别在培养后6、12、18、24h共4个时段于解剖显微镜下观察并记录晶状体混浊度的改变;检测晶状体水溶性蛋白、谷胱甘肽(GSH)的含量及总超氧化物歧化酶(T-SOD)和丙二醛(MDA)的活性。【结果】金钗石斛总生物碱和粗多糖均能减轻晶状体混浊度,并能显著升高晶状体水溶性蛋白、GSH含量及T-SOD活性,降低MDA的活性(均P<0.05),其中石斛总生物碱高剂量组效果最佳。【结论】金钗石斛总生物碱和粗多糖在体外均有一定的抗白内障作用,其机制与拮抗晶状体的氧化损伤有关,而总生物碱的效果优于粗多糖。Objective To probe into the inhibitory action of the total alkaloids and polysaccharides extracted from Dendrobium Mobile Lindl. on cataract in vitro. Methods The total alkaloids and polysaccharides were extracted from Dendrobium Mobile Lindl. by the chemical-dissolvent method. Lenses of Wistar rats were cultured in DMEM medium with H2O2 and total alkaloids or polysaccharides added. After culturing for 6, 12, 18, and 24 hours, the opacity of each lens was observed under an anatomical microscope, and the content of water-soluble protein, glutathione (GSH), and the activity of malonyldialdehyde (MDA) and total superoxide dismutase (TSOD) in each lens were measured with the kits respectively. Results The lens opacity can be reduced obviously by the total alkaloids and polysaccharides. The contents of water-soluble protein and GSH , and the activity of T-SOD in the lens were up-regulated, and the activity of MDA was degraded by the two extracts (P 〈 0.05). The above effects were obvious in the high-dose total alkaloids group. Conclusion Both of the total alkaloids and polysaccharides of Dendrobium Mobile Lindl. can inhibit cataract in vitro, and the total alkaloids have a better effect. Their mechanism may be related with the counteraction of oxidative damage.
