摘 要:目的探讨三七总皂甙(Panax notoginseng saponins,PNS)抗肾小管上皮细胞转分化(EMT)的分子病理机制。方法将雄性SD大鼠80只随机等分为4组:假手术组、UUO模型组、PNS治疗组和氯沙坦(ARB)治疗组。术后3、7、14天和21天每组随机选择5只大鼠处死,收集肾组织。分别用Western杂交、免疫组化检测α-SMA的表达;用Real-Time PCR检测UUO术后大鼠α-SMA mRNA的表达。结果大鼠肾组织α-SMA表达,对于3d、7d,仅UUO组、PMS组均高于SOR组,其余任意两组比较均相近;14d、21d,仅PNS组与氯沙坦组均相近,其余任意两组比较均不同。UUO组、PNS组、氯沙坦组的时间与α-SMA表达间存在相似的正相关的曲线关系。大鼠肾组织α-SMA mRNAΔCT值,SOR组、氯沙坦组的7d与14d观察值均相近,其他组14d观察值均低于7d。结论PNS能有效下调UUO大鼠肾脏组织α-SMAmRNA及其蛋白的表达,具有抗肾小管上皮细胞转分化的作用。Objective To reveal the effect of Panax notoginsenosides (PNS) improved unilateral ureteral obstruction (UUO) induced transdifferentiation of renal epithelial cells. Methods 80 male SD rats were randomly allocated into four groups, including sham- operation group (group 1), model group (group 2), PNS treated group (group 3),and losartan treated group(group 4). Group 3 and Group 4 were treated with PNS and losartan, respectivly. The other groups were administrated with water. At 3, 7, 14, 21 days after operation, 5 rats randomly selected from each group were killed. The expression of α-SMA in nephridialtissue were detected with immunohistochemitry, western blot and RT-PCR. Results The level of α-SMA in group 2 were obviously higher than that in other groups (P〈0.05). Compared with group 2, the level of α- SMA of group 3 was significantly low (P〈0.05). Conclusions PNS can downregulate the expression of α-SMA.
