摘 要:目的研究极低频正弦磁场对F0F1-ATPases活力的影响及其机制。方法强度为0.1—0.5mT、频率为4.7~96.0Hz的磁场分别处理从光合细菌(Rhodospirillum rubrum)中提取的细菌色素体(chromatophores)外膜上的F0F1-ATPases。结果0.5mT,4.7、12.0、60.0、72.0、84.0、96.0Hz磁场对F0F1-ATPases水解起促进作用,0.5mT、24.0Hz磁场对F0F1-ATPases水解起抑制作用,差异均有统计学意义(P〈0.05);0.3mT,4.7、24.0、60.0Hz的磁场对F0F1-ATPases仍有明显影响,但0.1mT的磁场则对F0F1-ATPases无明显影响。0.5mT,24.0、60.0Hz的磁场对二环己基碳二亚胺(DCCD)抑制的F0F1-ATPases仍分别有明显地抑制和促进酶活力的影响,差异有统计学意义(P〈0.05)。结论影响F0F1-ATPases活力的极低频磁场具有频率窗口,强度阈值在0.1~0-3mT之间;F0F1-ATPases,特别是F1是磁场作用的一个重要的位点。Objective To study the effects of extremely low frequency sinusoidal magnetic fields on hydrolysis of F0F1-ATPase and its mechanism. Methods The F0F1-ATPases which was localized on the outer surface of chromatophores were prepared from the cells of Rhodospirillum rubrum and were exposed to 0.1-0.5 mT,4.7-96.0 Hz magnetic fields. Results The hydrolysis activity of F0F1-ATPase was stimulated by 0.5 mT, 4.7,12.0,60.0,72.0,84.0 and 96.0 Hz magnetic fields respectively and inhibited by 0.5 mT, 24.0 Hz magnetic field(P〈0.05) ;0.3 mT,4.7,24.0 and 60.0 Hz magnetic fields also distinctly affected F0F1-ATPases activity respectively (P〈0.05),whereas 0.1 mT exposure caused no significant changes on that activity. When the hydrolysis activity of the F0F1-ATPases was inactivated by its inhibitor DCCD, the 0.5 mT, 24.0 Hz magnetic field still inhibited the hydrolysis activity of the F0F1-ATPase and 0.5 mT, 60.0 Hz magnetic field also had stimulating effects(P〈0.05 ). Conclusion The effects of magnetic fields on the hydrolysis activity of the F0F1-ATPases depend on not only magnetic frequency but also magnetic intensity. The threshold of magnetic intensity is between 0.1 mT and 0.3 mT. F0F1-ATPases, especially Fl-portion may be an end-point of magnetic fields.
