摘  要:【目的】优化百部总生物碱的提取工艺,为其在农业、医药、食品等方面的开发提供理论参数。【方法】在单因素试验的基础上,选取百部总生物碱提取温度、提取时间和乙醇质量分数3个因素进行Box-Benhnken中心组合设计,利用响应面分析法对其提取工艺参数进行优化。【结果】在提取温度57.76℃,提取时间13.18 h,乙醇质量分数75.21%时,百部总生物碱的理论含量最高,可达9.76 mg/g。根据实际试验情况,将其修正为提取温度57.8℃,提取时间13.2 h,乙醇质量分数75.2%,经验证此条件下百部总生物碱含量为9.63 mg/g,与理论值较为接近。【结论】修正后的百部总生物碱提取工艺优化参数准确可靠,有一定的实用价值。[Objective] The extraction technique of total alkaloid from Radix Sternona was optimized to provide theoretical parameters for its exploiture in agriculture,medicament, foodstuff and so on. [Method] On the basis of single-factor experiments, the effects of different temperature, time and ethanol weight percent on the extraction yield of total alkaloid from Radix Sternona were investigated with a three-factor BoxBehnken center-united design. The extraction parameters were optimized with Response Surface Methodology. [Result] The results indicated that the conditions of 57.76 ℃ ,75.21% ethanol and heating and refluxing for 13.18 h were optimum, and the yield of total alkaloid could be up to 9.76 mg/g. According to conditions of actual experiment,the optimum extraction parameters were modified at 57.8℃ for 13.2 h with ethanol weight percent 75.2%. The validated experiment showed the yield of total alkaloid was 9.63 mg/g under those conditions which was in consistency with the theoretic value. [Conclusion] The modified extraction parameters were credible and had some practical value.
