摘  要:目的:研究SD大鼠牙槽骨成骨细胞在不同压力刺激下环氧合酶-2(COX-2)蛋白的表达特点。方法:采用自行研制的可调压式细胞培育箱对原代培养的牙槽骨成骨细胞加载持续性静压力和间歇性静压力,用免疫细胞化学染色和Western blot检测COX-2蛋白的表达。结果:COX-2蛋白的表达强度随着压力刺激作用时间的延长逐渐增强,但两种不同压力刺激对其表达量无显著性差异。结论:一定范围内不同形式的压力刺激均可诱导大鼠牙槽骨成骨细胞COX-2蛋白表达,并继而影响牙槽骨成骨细胞的增殖、分化、成熟,继而影响牙槽骨的吸收及新生和重建。Objective:To determine the effects of different compressive pressures on inducible cyclooxygenase-2 expressions of cultured rat alveolar osteoblasts. Method: A self-made compressive device was used to apply continuously or intermittently compressive pressures on the monolayer cells. Immunocytochemical staining and immunoblots were performed with polyclonal rabbit anti-cyclooxygenase-2. Result: The expressions of cyclooxygenase-2 was increased in the manner of mechanical loading time, but showed no statistical difference between continuously and intermittently compressive pressure group. Conclusion: A certain range of different mechanical stimulations can induce the expressions of cyclooxygenase-2 in cultured alveolar osteoblasts and affect the biological characters of alveolar osteoblasts such as proliferations, differentiations and maturities and then lead to alveolar absorptions, reproductions and rebuildings.
