摘  要:目的:了解HIV/AIDS感染者进行CD4+T淋巴细胞检测时随着血样采集后放置时间的延长,CD4+T淋巴细胞计数的变化。方法:用FACSCount流式细胞仪对我市HIV/AIDS病人送检的30份血样采集后分别放置8、24、48、72、96 h进行荧光染色,染色后放置30 min进行CD4+T淋巴细胞计数。并将结果进行平均值(Mean)、标准差(SD)和变异系数(CV)的统计计算。结果:同一样本放置8~96 h检测的数据差异<20%,为可接受范围。其中同一样本采集后放置不同时间检测的30份血液中有20份5次重复测定值的CV值<5%,占66.7%。相关性分析显示:五个时间点上的检测结果相互之间都具有很好的相关性(P<0.001),且相关系数(r值)均≥0.99。结论:在实验室其它条件(温度、试剂等)不变的情况下,用FACSCount流式细胞仪检测CD4+T淋巴细胞,血样采集后放置时间可延长至96 h。Objective: To define the influence on CD4^+T - cell counts for prolonged stored time after blood collection. Methods:30 blood samples were collected for this study, CD4^+T - cell counts were determined at 8, 24, 48, 72 or 96 hours after blood collection. Mean, SD and CV values were calculated and compared for the data from 5 different time - points, respectively. Results:CD4^+ T - cell counts from all samples showed less than 20% coefficient variation among data from 5 different time -points after blood collection. Among 30 samples, 20 (66.7%) showed 〈 5% coefficient variation for samples stored up to 96 hours after blood collection, respectively. And correlation analysis showed, the data from different time - points highly correlate with each other (P 〈 0. 001, r≥0. 99). Conclusion:With samples stored at room temperature and matched reagents, CD4^+ T - cell counts may be reliably quantified with FACSCount on samples stored for up to 96 hours after blood collection.
