摘  要:目的:建立大黄酸在小鼠后肢骨中药物浓度快速HPLC测定方法,初步考察大黄酸的趋骨性。方法:后肢骨样品经冷冻、粉碎、匀浆、去蛋白和有机溶剂提取处理后,RP-HPLC进行分析。采用Phenomenex ODS-C18(250mm×4.6mm,5μm)色谱柱;流动相为甲醇-乙酸水溶液-三乙胺(400:150:0.55,乙酸调pH5.0);流速1.0mL·min^-1;柱温25℃;检测波长为420nm。结果:后肢骨中大黄酸的方法回收率在98%~106%,在0.04~20.00μg·g^-1范围内线性关系良好,相关系数为0.9999,日内、日间RSD均〈6%,重现性良好。结论:本法简便、准确、灵敏,适用于大黄酸在小鼠骨内的药物分析。大黄酸具有较高的趋骨性。Objective :To establish a RP-HPLC method for the determination of rhein level in bone and observe its bone-targeted character. Methods:After refrigeration, fragmentation, precipitation protein and extraction, the concentration of rhein was analyzed by RP-HPLC. The separation was performed on a Phenomenex ODS-C18 column (250 mm × 4.6 mm,5 μm) with mobile phase of methanol-acetic acid water solution-triethylamine (400: 150: 0.55 ,pH 5.0). The UV-deteetor was set at 420 nm. Results:The recoveries of rhein in bone were more than 98% and less than 106%. The calibration curve was linear in the range of 0.04 - 20.00 μg·g^-1 with a correlation coefficient of 0. 999 9. The RSDs of intra-day and inter-day were less than 6%. Conclusion:The method is simple, precise,accurate and can be used for studies of rhein in mouse bone. And rhein has the trend of bone affinity.
