固定化硫酸多粘菌素体外抗微生物效果的研究

摘　　要：目的观察固定化硫酸多粘菌素体外抗微生物的效果。方法用戊二醛固定法将硫酸多粘菌素固定在玻璃纤维滤纸上，采用改良振荡瓶法进行抗菌效果检测。结果用pH9～10碱性缓冲液稀释制备硫酸多粘菌素固定化玻璃纤维滤纸，染菌后在室温下24h对大肠杆菌和金黄色葡萄球菌平均抑菌率均达到99．99％。固定化多粘菌素含量为1mg／ml玻璃纤维滤纸染菌后置室温下24h，对大肠杆菌和金黄色葡萄球菌抑菌率均达到99．99％；在多粘菌素浓度为5mg／ml条件下对枯草杆菌黑色变种芽孢抑菌率不达标；多粘菌素浓度达到2．5mg／ml条件下，对f2噬菌体抑制率达到99．9％以上。固定化多粘菌素玻璃纤维滤纸经37℃储存3个月，对4种试验微生物抑制作用基本没有下降。结论固定化硫酸多粘菌素玻璃纤维滤纸具有较好的抗菌作用，储存性能稳定。Objective To observe the antimicrobial activity of immobilized polymyxin sulfate in vitro. Methods Polymyxin sulfate was immobilized on glass fiber filter paper by glutaraldehyde immobilization method, and antimicrobial activity was determined by means of modified shaking flask method. Results At room temperature the average inhibiting rates of Escherichia coli and Staphylococcus aureus exposed to the filter paper containing immobilized polymyxin sulfate diluted by pH 9 - 10 basic buffer for 24 h were 99.99% ; those ofE. coli and S. aureus exposed to the glassfiber filter paper contai- ning immobilized polymyxin sulfate at the concentration of 1 mg/ml for 24 h attained 99.99% , but immobilized polymyxin sulfate at the concentration of 5 mg/ml could not inhibit Bacillus subtilis var. niger spores. The inhibiting rate of E. coli phage f2 exposed to polymyxin sulfate at the concentration of 2.5 mg/ml attained over 99.9%. The efficacy of glass fiber filter paper with immobilized polymyxin sulfate in inhibiting the four kinds of representative microorganisms essentially did not decrease after storing at 37~C for 3 months. Conclusion Glass fiber filter paper with immobilized polymyxin sulfate has not only excellent antlmicrobial activity, but also good storage stability.
