摘  要:背景:甲状旁腺激素是骨代谢的重要调节因子,膜型基质金属蛋白酶1在骨吸收的调控中起重要作用。目的:观察甲状旁腺激素对人成骨样MG-63细胞膜型基质金属蛋白酶1表达的影响。设计、时间及地点:观察对比实验,于2001-03/2002-07在中南大学湘雅二医院代谢内分泌研究所实验室完成。材料:人成骨样MG-63细胞株,甲状旁腺激素(1~34)。方法:分别应用1,10,100nmol/L的甲状旁腺激素(1~34)干预MG-63细胞24h;或分别用10nmol/L甲状旁腺激素(1~34)干预MG-63细胞2,6,24,48h。每干预组设3个平行样本,重复3次。主要观察指标:采用Northern杂交法检测膜型基质金属蛋白酶1 mRNA表达水平;Western杂交法检测膜型基质金属蛋白酶1蛋白表达水平。结果:①1,10,100nmol/L的甲状旁腺激素(1~34)对MG-63细胞膜型基质金属蛋白酶1 mRNA及蛋白的表达有抑制作用,呈剂量依赖性(P<0.05)。②10nmol/L的甲状旁腺激素(1~34)在干预2~48h内对膜型基质金属蛋白酶1 mRNA及蛋白的表达有抑制作用,呈时间依赖性(P<0.05)。结论:甲状旁腺激素具有抑制成骨样细胞膜型基质金属蛋白酶1表达的作用,此可能为甲状旁腺激素诱导骨吸收的作用途径之一。BACKGROUND: Parathyroid hormone is an important regulatory factor in the bone metabolism, and membrane-type matrix metalloproteinase-1 plays an important role in the regulation of bone resorption. OBJECTIVE: To observe the effects of parathyroid hormone on the expression of membrane-type matrix metalloproteinase-1 in human osteoblast-like MG-63 cells. DESIGN, TIME AND SETTING: Contrast observation was conducted at the laboratory of Institute of Endocrinology & Metabolism, the Second Xiangya Hospital of Central South University between March 2001 and July 2002. MATERIALS: Human osteoblast-like MG-63 cell line; parathyroid hormone (1-34). METHODS: MG-63 cells were treated with 1, 10, 100 nmol/L parathyroid hormone (1-34) for 24 hours, respectively, or were treated with 10 nmol/L parathyroid hormone (1-34) for 2, 6, 24, 48 hours, respectively. Each intervention group had three parallel samples and each intervention was repeated for three times. MAIN OUTCOME MEASURES: Northern Blotting were used to determine the expression level of membrane-type matrix metalloproteinase-1 mRNA; Western Blotting were performed to detect the protein expression of membrane-type matrix metalloproteinase-1. RESULTS: Our study showed that 1, 10, 100 nmol/L parathyroid hormone (1 34) inhibited membrane-type matrix metalloproteinase-1 mRNA and protein expression in a dose-dependent manner in MG-63 cells (P 〈 0.05). Within 2-48 hours of intervention, 10 nmol/L parathyroid hormone (1-34) inhibited membrane-type matrix metaLloproteinase-1 mRNA and protein expression in a time-dependent manner in MG-63 cells (P 〈 0.05). CONCLUSION: Parathyroid hormone can inhibit the expression of membrane-type matrix metalloproteinase-1 in osteoblast-like MG-63 ceils, it may be one of the ways with which parathyroid hormone induces bone resorption.
