摘 要:目的观察益气活血法对脑出血大鼠脑内损伤区凝血酶敏感蛋白-2(TSP-2)的影响,探讨益气活血法治疗脑出血的机制。方法SD大鼠随机分为正常组、假手术组、模型组、益气活血组、益气组和活血组;采用Ⅶ型胶原酶诱导大鼠脑出血模型,再对相应的组分别灌服补阳还五汤及该方的益气药和活血药,在不同的时间点取脑组织并用免疫印迹分析鼠脑TSP-2的表达。结果正常组和假手术组不同时间点TSP-2表达无明显变化;模型组TSP-2在脑出血后1d即开始表达,1~7d各时间点表达无明显差异,7~28d表达呈逐渐上升趋势,并在28d达高峰;益气活血组TSP-2表达14~21d时上升趋势较其它三组缓慢,在21d表达较其它三组弱,在28d表达较其它三组强。结论益气活血法有可能通过影响脑出血后损伤区TSP-2的表达,在受损脑组织进行血管重建的阶段,抑制TSP-2的表达,降低其对血管新生的抑制作用,延长血管新生的时间,并在血管重建后期上调TSP-2表达,抑制血管过度增殖,以保证整个血管新生过程的顺利和效率。Objective: To investigate the effects of Yiqihuoxue therapy on the expressions of TSP - 2 in brains of Intracerebral Hemorrhage (ICH) rats and explore its mechanisms. Methods: One hundred and fifty -five SD rats were randomly divided into six groups including normal control gorup, sham operated group, ICH model group, Yiqihuoxue therapy group, Yiqi therapy group and Huoxue therapy gorup. By injecting collagenase type VII stereotaxically into right globus, ICH model was made. Yiqi therapy gorup, Huoxue therapy group and Yiqihuoxue therapy group were respectively administered at Yiqi decoction, Huoxue decoction and Buyanghuanwu decoction. Extractd brain tissue at different time points and used Western blotting to examine the expressions of TSP - 2 in rat brains. Results: Studies showed that the expressions of TSP - 2 in normal control group and sham operated group had no significant changes at different time points. The expressions of TSP - 2 in ICH model group could be observed at 1st day and had no sig- nificant changes at different time poltns from 1st day to 7th day. From 7th day to 28th day it increased gradually till reached peak at 28th day. The expression of TSP - 2 in Yiqihuoxue therapy group increased slower than that in other three groups from 14th day to 21st day. The expression of TSP -2 Yiqihuoxue therapy group was lower than that in model group at 21th day but was higher than that in model group at 28th day. Conclusion: Yiqihuoxue therapy can diminish the expressions of TSP - 2 in ICH rat brains at the stage of angiogenesis to degrade the depreasant effects on angiogenesis and prolong the period of angiogenesis. At the anaphase of angiogenesis it can also up -regulate the expressions of TSP - 2 to inhibit hyperplasy of newborn vessels then ensure angiogenesis can proceed smoonthly and effectively.
