摘 要:目的探讨葛根素对自发性高血压大鼠核因子-kappaB(NF-κB)表达的影响。方法将24只自发性高血压大鼠随机分为对照组、葛根素组(腹腔注射葛根素)和贝那普利组(灌胃贝那普利)。免疫组化染色法观察各组肾血管NF-κB的表达,放免法检测各组大鼠血清血管紧张素Ⅱ;光镜下观察各组大鼠肾动脉狭窄的程度。结果NF-κB在自发性高血压大鼠肾血管均有表达;与对照组比较,葛根素组和贝那普利组NF-κB表达降低,血管紧张素Ⅱ下降,肾动脉狭窄程度明显减轻。结论葛根素可能通过抑制血管紧张素Ⅱ引起的NF-κB活化以下调NF-κB表达,从而减轻肾动脉狭窄程度。Objective: To explore the effects of Puerarin on NF-κB expression in kidneys of spontaneous hypertensive rats. Methods: 24 spontaneous hypertensive rats(SHR) were divided into three groups; control group, Purearin group(intraperitoneal injection) and benazepril group (intragastric administration) . Renal NF -κB expression in SHR were studied by immunocyto chemical techniques, angiotension Ⅱ in every group rat' s serum were assayed by radiation immunological test. Renal artery stricture of every group rat were obseryed under optic microscope. Results: NF -κB was expressed in spontaneous hypertensive rats; as compared with control group, NF -κBexpression decreased remarkably in benazepril group and Puerarin group. And as compared with control group, angiotension Ⅱ expression was downregulated in benazepril gorup and Puerarin group with statistical difference, and renal artery stricture in the two groups was alleviateded distinctly. Conclusion: Puerarin alleviateds renal artery stricture by the way it inhibits angiotension Ⅱ expression which actives NF -κB.
