摘 要:[目的]研究2005~2006年河北省分离的H1N1型流感病毒毒株HA1基因特性。阐明HA1基因的变异与流感流行的关系。[方法]用MDCK分离培养流感病毒,提取病毒核糖核酸(RNA),采用RT-PCR扩增病毒HA1基因,纯化产物进行核苷酸序列测定,用DNAStar软件作分析处理。[结果]2005~2006年在河北省流行的H1N1型流感病毒HA1基因核苷酸和氨基酸序列与同时期的H1N1亚型疫苗株A/NewCaledonia/20/1999相比已发生较大变异,核苷酸同源性为97.0%~98.1%,氨基酸同源性为97.9%~98.7%,有5~7个氨基酸发生了替换,但均不在抗原表位。种系进化结果显示A/河北南市/37/2005与A/河北新市/56/2005和A/河北新市/129/2006位于2个小分支,均距A/NewCaledonia/20/1999较远,A/河北南市/37/2005与A/NewYork/221/2003有较近的亲缘关系。[结论]H1N1型流感病毒HA1基因已发生明显变异,但对抗原性改变意义不大;近几年H1N1持续低流行使人群易感性得以积累是今年H1N1型流感流行的主要原因;2005~2006年河北省同时流行着至少2支种系发生距离较远的H1N1型流感病毒。[Objective] To study the characteristics of HA1 gene isolated from influenza virus H1N1 subtype in Hebei from 2005 to 2006,and to clarity the relationship between the influenza epidemiology and the variations of HA1 virus.Methods The viruses were isolated and propagated by MDCK cells.RNAs were extracted and HA1 genes were amplified by reverse transcription polymerase chain reaction(RT-PCR).The purified PCR products were sequenced.The data obtained were analyzed with the software DNAStar.Results The influenza viruses H1N1 subtype in HeBei from 2005-2006 was significantly different from the A/New Caledonia/20/1999 which was the vaccine strain recommend by WHO at that time in HA1 region,the homology of nucleotide was at the range of 97.0% to 98.1%,and the homology of amino acide was at the range of 97.9% to 98.7%.5 to 7 amino acid replaced but not occurred in epitope.The results of germ line evolution demonstrated that the A/Nanshi of Henan/37/2005 was located in two branch with A/Xinshi of Hebei/56/2005 and A/Xinshi of Hebei/129/2005,but far from the A/New Caledonia/20/1999.A/Nanshi of Henan/37/2005 had closely genetic relationship with A/New York/221/2003.Conclusion The HA1 gene of influenza H1N1 viruses shows obvious variation,which has little significance on the change of antigen;The main reason for the prevalence of influenza H1N1 viruses is the continued low popularity of HINI which cause the susceptibility of herd.Two types of germ lines with long relationship with each other have occurred popularity in Hebei from 2005 to 2006.
