摘 要:目的:通过比较不同时期正常胎鼠和地塞米松诱导的腭裂胎鼠中间嵴上皮细胞(MEE)和腭突间充质细胞(EPM)的数目和分布情况,初步探讨腭裂形成的机制。方法:选取孕鼠30只,随机分为对照组和实验组,实验组于妊娠GD11.5腹腔内注射50mg/kg地塞米松,干扰胎鼠腭突发育,分别于GD12.5,13.5,14.5,15.5,16.5时取材,共获胚胎284只,取头部标本做冠状切片,切片厚6μm,HE染色,光镜下观察,记录MEE细胞的数量和EPM细胞的数目和分布情况。结果:实验组和对照组腭突突部EPM明显多于根部,且实验组EPM发育滞后于对照组;GD13.5,14.5,15.5时实验组EPM细胞数量少于对照组;实验组MEE细胞在GD15.5,16.5时大量存在,而对照组GD14.5时开始融合甚至消失。结论:地塞米松导致的EPM细胞增殖抑制和MEE细胞的转归异常,是腭裂形成的重要原因。Objective:To investigate the primary mechanism of palate clefts. The distribution and numbers of Embryonic Palatal Mesenchymal Cells (EPMs)and medial edge epithelial cells (MEEs)on palatine process between normal and palate cleft mouse fetus were contrasted by Dexamethasone on different periods. Methods..30 pregnant mice were randomly divided into control group and experiment group. The mice in experi-ment group were given Dexamethasone,50mg/kg by intraperitoneal injection on GD11.5, then killed by cervical dislocation on GD12. 5,13. 5,14. 5,15. 5,16. 5,and 284 embryo were got. Heads were embeded by paraffin on coronal plane,sectioned 6μm and HE stained. Then observed by microscope and the numbers of EPMs and MEEs in different groups were recorded. Results:The numbers of EPMs on tip of palatine process were more than that of palatine process root. The development of EPMs in experiment groups was lower than that of con- trol group. The numbers of experiment group EPMs were less than that of control group on GD13. 5,14. 5, 15.5. Many MEEs were distributed on the edge of palatine process on GD15. 5,16. 5, but in control group MEEs began to fuse on GD14.5 ,even disappeared. Conelusions: Dexamethasone can inhibite the proliferation of EPMs and abnormal turnover of MEEs,which is an important cause for the development of cleft palate.
