法氏囊三肽囊素(Bursin)的研究Ⅰ.抗 Bursin 单克隆抗体杂交瘤细胞株的建立

摘　　要：以戊二醛法制备ＢｕｒｓｉｎＫＬＨ，免疫ＢＡＬＢ／ｃ小鼠，取脾细胞，以ＰＥＧ为融合剂，与ＳＰ２融合，用间接ＥＬＩＳＡ法筛选出与Ｂｕｒｓｉｎ－ＢＳＡ结合、不与ＢＳＡ结合的２Ｆ９－４克隆株，经鉴定，抗体属性为ＩｇＭ，Ｋ轻链。水溶性Ｂｕｒｓｉｎ可阻抑２Ｆ９－４对鸡法氏囊的反应，切片标本经ＰＢＳ处理，反应性消失，免疫组织化学染色于法氏囊上获得特征性阳性结果。Synthetic bursin (Lys-His-Gly-NH 2) was coupled to keyhole limpet haemocyanin (KLH) via glutaraldehyde. BALB /c mice were immunized with the KLH-bursin conjugat, then the spleen cells of immunized mice were fused with SP 2 cells. By detecting the cell culture with indirect ELISA, one hybridoma cell strain (2F9-4 strain) was obtained, the monoclona antibodies (McAb) to KLH-bursin, produced by this strain, were reactive with bursin-BSA (bovine serum by albumin) by indirect ELISA, and were unreactive with BSA. The McAb belonged to K L chain IgM. The McAb intensely and specifically stained some kinds of bursal epithelial cells by immunohistochemistry, the McAb absorbed with bursin failed to stain the bursal epithelial cells, interestingly, no stain was found by immunohistochemistry.
