摘 要:目的了解糖尿病(diabetes mellitus,DM)大鼠脑内血管紧张素Ⅱ(AngⅡ)含量及其1型受体(AT1)表达的改变。方法以链脲佐菌素(STZ)诱导的DM大鼠为研究对象。采用免疫组化观测大鼠脑内AT1的表达;用放射免疫分析法测定血浆及脑组织AngⅡ的含量。结果DM发生14天后,血浆和下丘脑的AngⅡ含量升高,延髓AngⅡ含量降低,视上核(SON)、室旁核(PVN)、孤束核(NST)的AT1表达显著增强,而穹隆下器官(SFO)的AT1表达则显著减弱。结论DM大鼠的中枢和血浆的AngⅡ含量及AT1在脑内的表达有异常改变。Objective To assess the changes of brain-local angiotensin Ⅱ (Ang Ⅱ ) and the expression of Ang Ⅱ type 1 receptor (ATI) in diabetic rats. Methods Male Wistar rats were allocated to three groups: control group,non-diabetic rats; DM group, STZ-indueed diabetic rats. After 14 days, the rats were sacrificed and the various components of RAS were determined. Immunohistoehemistry and computer software for image analysis were used to evaluate the expression of AT1. The level of Ang Ⅱ in plasma, hypothalamus and medulla oblongata was measured by radioimmunoassay (RIA). Blood glucose, body weight; drinking volume per day were measured concurrently. Results Compared with control group, DM rats were associated with enhanced Ang Ⅱ level in plasma, hypothalamus and a decreased Ang Ⅱ level in medulla oblongata. The expression of AT1 was significantly upregulated in the supraoptie nucleus (SON), paraventrieular nucleus (PVN) and nucleus of solitary tract (NST), whereas downregulated in subfomieal organ (SFO). Diabetic rats developed hyperglycemia, as well as polydipsia and polyuria, and failed to gain weight. Conclusions There are changes with the level of Ang Ⅱ in brain, plasma, and also the expression of brain local AT1 in DM rats.
