摘 要:为研究红花注射液对大鼠溃疡性结肠炎组织损伤的治疗作用及其机制,选取40只SD大鼠随机分为4组:正常组、模型组、红花注射液组和地塞米松组。用三硝基苯磺酸(TNBS)灌肠建立溃疡性结肠炎SD大鼠动物模型。腹腔注射用药2周后,评价大鼠疾病活动指数(DAD及组织学损伤评分,检测结肠组织中过氧化物酶(MPO)活性和肿瘤坏死因子-α(TNF—α)、白细胞介素-8(IL-8)的表达。结果显示,与模型组相比,红花注射液腹腔注射给药能降低模型大鼠DAI[(2.62±0.47)vs(3.15±0.39),P〈0.01]、组织学评分[(3.32±0.78)vs(5.26±0.73),P〈0.01]和MPO活性[(0.78±0.07)U/g vs(1.41±0.09)U/g,P〈0.01],抑制TNF-α[(30.16±7.78)vs(54.12±9.23),P〈0.01]、IL-8E(35.46±7.67)vs(59.44±8.64),P〈0.01]的表达。结果表明,红花注射液可以改善模型大鼠DAI及组织学评分,降低MPO活性,从而对大鼠溃疡性结肠炎起到治疗作用。抑制TNF—α、IL-8的表达可能是其治疗机制之一。The objective of this study is to probe into the therapeutic efficacy and mechanism of carthamus tinctorius injection in treating the experimental colitis in rats. Trinitrobenzene sulfonic acid enema was applied to induce ulcerative colitis in SD rats(rats model),and 40 rats were randomised into 4 groups:rats in group A were normal rats,rats in group B receiving no treatment,rats in group C receiving carthamus tinc torius injection treatment,and rats in group D receiving dexamethasone treatment. The disease activity in dex(DAI),score of colonic histology,activity of myeloperoxidase(MPO) and the expression of tumor necrosis factor-α(TNF-α) and interleukin-8(IL- 8) were then measured after two weeks of intraabdominal in jection of drugs. Compared with group B,the value of DAI in group C was decreased(2.62±0.47 vs 3.15±0.39, P 〈0.01) ;the score of colonic histology was increased(3.32±0.78 vs 5.26±0.73, P 〈0.01) ;the activity of MPO was increased(0. 78±0.07 u/g vs 1.41±0.09 U/g, P 〈0.01);and the expression of TNF-α and IL-8 was decreased(30.16±7.78 vs 54.12±9.23, P〈0.01;and 35.46±7.67 vs 59.44±8.64, P 〈0.01) respectively. It is concluded that carthamus tinctorius injection is of therapeutic efficacy in the treatment of ulcerative colitis in rats,and one of the mechanism is its inhibition of the expression of TNF-α and IL-8.
