白附片煎煮时间、给药剂量与温肾补脾功效的相关性研究

摘　　要：目的:研究白附片煎煮时间、给药剂量与温肾补脾功效的相关性,确定药物发挥温肾阳和脾阳功效的最佳煎煮时间和给药剂量。方法:根据附子补火助阳的功效,选择肾阳虚模型、阳虚便秘模型等,运用均匀设计和回归分析方法,确定药物不同功效的最佳煎煮时间和给药剂量。结果:白附片能明显改善肾阳虚小鼠一般状态,显著延长低温游泳衰竭时间和爬杆时间;显著缩短阳虚便秘小鼠排便潜伏期,增加排便颗粒数,能明显促进胃肠蠕动,提高胃肠推进率。白附片发挥温肾阳作用的最佳煎煮时间为6h,最佳给药剂量为12g/kg;温阳通便药物的最佳煎煮时间为6h,最佳给药剂量为12g/kg。结论:白附片具有显著的温补脾肾阳气作用,煎煮时间、给药剂量与温阳功效显著相关。发挥温肾补脾功效的最佳煎煮时间和给药剂量为6h12g/kg。Objective： To find out the best decoction time （DT） and administration dose （AD） of the WPLRA acting, the correlation was investigated between DT, AD and efficacy of warming kidney and spleen Yang of White Prepared Lateral Root of Aconite （WPLRA）. Methods： In accordance with the efficacy of the WPLRA supplementing fire and strengthening Yang, several animal models, including a constipation induced by insufficiency of Yang, kidney Yang deficient models, Uniform Design （UD） and Regression Analysis were applied to confirming the best DT and AD of the different actions. Results： The WPLRA evidently improved general state of the kidney-Yang deficiency mice, remarkably extended their swimming depleted and climbing time in lower temperature. The WPLRA also markedly decreased the defecation latency, promoted defecation amount, and improved peristalsis frequency of intestines and stomach. The best DT and AD for the WPLRA to warm the kidney Yang and spleen Yang to smooth defecation were 6 hours and 12g/kg respectively. Conclusions ： The WPLRA possesses the remarkable efficacy of warming yang. The action of warming Yang is closely correlated with the DT and AD. The best DT and AD for the WPLRA to warm kidney and spleen Yang are 6 hours and 12g/kg respectively.
