摘 要:目的将分子烙印聚合物(MIP)作为固相萃取的吸附剂,研究其分子识别性能。方法采用分子烙印技术制备了美托洛尔分子烙印固相萃取柱,并与空白聚合物柱对照,探讨了其在非平衡吸附条件下的最大负载量,确定了上样、淋洗和洗脱的最佳萃取模型。结果美托洛尔的分子烙印固相萃取柱对原模板具有较高的选择性和识别能力,而空白聚合物却不具有这样的特性。在优化的萃取条件上,美托洛尔与其结构相似的卡维地洛和普萘洛尔在柱上得到了很好的分离。结论MIP作为固相萃取吸附剂为解决药物的纯化提供了一条可行的有效途径。OBJECTIVE To study the molecular recognition properties of the molecularly imprinted polymer(MIP) as the adsorbent of solid-phase extraction ( SPE). METHODS The SPE columns were filled with metoprolol-molecularly imprinted polymers which were prepared by adopting molecular imprinting technique. By contrast with the blank polymer, the maximum loading capacities of MIP under the non-equilibrium conditions were investigated and the optimum models concerning on the loading, washing and elution solution were founded. RESULTS The metoprolol-imprinted extraction column exhibited high selectivity and good recognition ability for the template, howerver the blank polymer didn't. Under the optimum extraction conditions, metoprolol and its structurally related compounds carvedilol and propranolol were separated primely on this kind of solid-phase column. CONCLUSION MIP, as the adsorbent of SPE, will provide an effective way to solve the purification of drugs.
