摘  要:目的探讨银杏叶提取物(EGb)诱导大鼠主动脉平滑肌细胞(SMC)中血红素氧合酶-1(HO-1)表达的量效关系,并初步探讨EGb诱导HO-1表达的分子机制。方法复苏并传代培养大鼠主动脉SMC,分组实验,分别采用0,50,100,200,400μg/ml 5个不同浓度的EGb以及200μg/ml EGb加放线菌素D(ActD)孵育SMC 8 h,Western blot法检测HO-1蛋白,以HO-1条带灰度值代表HO-1的表达量。结果不加EGb的对照组中HO-1蛋白仅有微量基础表达,加用EGb各组中HO-1蛋白的表达随EGb浓度的增加而增加(P<0.05),在EGb浓度为200μg/ml时达到峰值,随后则有所下降。加用ActD能显著抑制EGb所诱导的HO-1蛋白表达(P<0.05)。结论EGb能够诱导大鼠主动脉SMC中HO-1表达,且这种表达在一定范围内具有量效关系;EGb诱导HO-1表达有赖于基因重新转录。Objective To explore the dose - effect relationship and the molecular mechanism of the heine oxygenase - 1 ( HO - 1 ) expression induced by the extract of Gingko biloba (EGb) in rat aorta smooth muscle cells ( SMC ). Methods The SMC was revived,passage cultivated and divided into different groups. The cells in each group were incubated with 5 concentrations of EGb such as 0,50,100,200,400 μg/ml and EGb 200 μg/ml + ActD4μm respectively. The incubation lasted for 8 hours, then Western blot was applied to analyze HO - 1 protein expression. Results There was only basal microamount expression in the group without EGb. EGb significantly induced the expression of HO - 1 in SMC and the expression increased with the increase of EGb concentration (P 〈0.05),it reached the peak amplitude under the condition of EGb 200 μg/ml, then deseeded. EGb- induced HO - 1 expression was significantly inhibited by ActD. Conclusion EGb can induce HO - 1 expression in rat aorta SMC and the expression has a dose - dependent feature under a limited EGb concentration range. EGb - induce HO - 1 protein expression depends on HO - 1 genetic retranscription.
