摘  要:目的研究膜荚黄芪叶愈伤组织诱导、生长及黄芪多糖合成的最佳生长调节剂组合。方法以膜荚黄芪叶为外植体,运用正交设计研究6-BA、2,4-D和NAA等3种植物生长调节剂组合对愈伤组织诱导、培养及黄芪多糖合成的影响。结果分别以诱导率、相对生长速率和黄芪多糖含量为指标,结果表明NAA和6-BA对愈伤组织诱导率影响显著,6-BA、2,4-D和NAA都对愈伤组织的生长影响显著,2,4-D和NAA对黄芪多糖合成影响显著。结论诱导愈伤组织的最佳培养基为:MS+2,4-D(0.5 mg.L-1)+6-BA(1.0 mg.L-1)+NAA(2.0 mg.L-1);愈伤组织生长的最佳培养基为:MS+2,4-D(0.5 mg.L-1)+6-BA(2.0 mg.L-1)+NAA(2.0 mg.L-1);促进黄芪多糖合成的最佳培养基为:MS+2,4-D(1.0 mg.L-1)+6-BA(1.0 mg.L-1)+NAA(1.0 mg.L-1)。Objective To study the optimum combinations of plant growth regulators in the induction, growth and polysaccharide biosynthesis of callus from the leaves of Astragalus membranaceus ( Fisch. ) Bge. Methods Taking the leaves of Astragalus Membranaceus (Fisch.) Bge as explants, three kinds of plant growth regulators, including 6 - BA,2,4 - D and NAA, were introduced into callus induction and growth, and polysaccharide biosynthesis by orthogonal design. Results Three parameters, including induction rate, relative growth rate and content of Astragalus polysaccharide, were used as criteria of estimating the efficiency of three regulators, both NAA and 6 - BA could induce callus most effectively, three of 6 - BA, 2,4 - D and NAA all could affect the growth of callus remarkably, and both 2,4 - D and NAA could promote the synthesis of polysaccharides effectivelyas compared with 6 - BA. Conclusion The optimal culture medium for callus induction is:MS + 2,4 - D (0.5mg · L^ -1 ) + 6 - BA ( 1.0 mg · L^ -1 ) + NAA(2.0 mg· L^ -1 ) ;The optimal culture medium for callus growth is: MS +2,4 - D(0.5mg · L^ -1 ) + 6 - BA(2.0 mg · L^ -1 ) + NAA (2.0 mg · L^ -1 ) ; and the optimal culture medium for the synthesis of Astragalus polysaccharides is : MS + 2,4 - D (1.0mg· L^ -1) +6-BA(1.0mg· L^ -1) +NAA(1.0mg· L^ -1).
