摘  要:目的探讨玄参对心室重构大鼠血管紧张素Ⅱ(AngⅡ)及其1型受体基因表达(AT1A mRNA)的影响。方法采用甲状腺素致大鼠心室重构及腹主动脉不完全结扎致大鼠心室重构两种动物模型,观察玄参对心脏指数(heart weight index,HWI)、心肌血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、醛固酮(aldosterone,ALD)及血管紧张素Ⅱ受体1型基因(AT1A mRNA)表达水平的影响。结果与模型组相比,玄参可显著降低动物HWI,降低心肌AngⅡ,ALD含量,降低AT1A mRNA的过量表达。结论玄参对心室重构具有明显的改善作用,其机制可能与抑制AngⅡ,ALD生成和AT1A mRNA表达有关。Objective To investigate the influence of Xuanshen on the concentration of angiotensin Ⅱ ( Ang Ⅱ ) and the expression of angiotesin Ⅱ receptor ( Type 1A) mRNA ( AT1A mRNA) in ventricular remodeling rats. Methods Two ventricular remodeling models were established. One was induced by thyroxin and the other was induced by abdominal aortic stenosis in rats. The heart weight index (HWI) , the Ang Ⅱ and ALD concentration, and the AT1A mRNA expression were determined after a period of Xuanshen administration. Results Compared with those of ventricular remodeling rats, the HWI, the concentration of Ang Ⅱ and ALD and the expression of AT1A mRNA were decreased in groups treated with Xuanshen. Conclusion Xuanshen has an obviously effect on preventive ventricular remodeling. The mechanism may be related to the decline of Ang Ⅱ and ALD concentration and the inhibition of AT1A mRNA expression.
