摘  要:目的探讨四物汤对环磷酰胺所致贫血小鼠血清促红细胞生成素(EPO)及肾EPO mRNA表达的影响。方法用环磷酰胺腹腔注射建立小鼠贫血模型,同时给小鼠灌服四物汤,ELISA方法检测小鼠血清EPO的含量;Trizol试剂提取肾组织总RNA,反转录-聚合酶链反应(RT-PCR)方法检测肾组织EPO mRNA表达情况(β-actin为内对照标准)。结果四物汤能显著升高小鼠血清EPO水平,明显促进肾组织EPO mRNA的表达。结论四物汤能抑制环磷酰胺所致的小鼠贫血,其作用机制可能是通过促进肾组织EPO mRNA的表达,使肾脏合成EPO增多,从而促进红系细胞生长。Objective To investigate the effect of Siwu Tang on erythropoietin(EPO) and EPO mRNA expression of Anemia induced by cyclophamide in mice. Methods Anemia was induced by cyclophamide in mice,and Siwu Tang were administered, Serum EPO level was measured by ELISA. Total RNA was isolated by Tfizol reagent. The mRNA expression of EPO in mouse kidney was measured by RT -PCR( β -actin gene as a control). Results Siwu Tang could significantly increase serum EPO level and promote the mRNA expression of EPO in kidney. Conclusion Siwu Tang has inhibitory effects on Cyclophamide - induced anemia in mice and the mechanism may be related to increasing EPO production by promoting the mRNA expression of EPO in kidney, and stimulating the growth of erythroid cells.
