摘 要:目的探讨丹参酮ⅡA(TanⅡA)对缺血再灌注损伤大鼠保护作用及对额顶部皮质中性粒细胞浸润、自由基含量、能量代谢的影响。方法将大鼠随机分为假手术组、缺血再灌注组、TanⅡA低剂量治疗组和TanⅡA高剂量治疗组,线栓法建立局灶性脑缺血再灌注模型。TanⅡA高、低剂量治疗组于术前连续灌胃给药3d,1次/d。用生化法观察缺血90min再灌注24h大鼠额顶部皮质髓过氧化物酶(MPO)活性、超氧化物歧化酶(SOD)、丙二醛(MDA)含量、三磷酸腺苷(ATP)含量及脑含水量变化,并进行2,3,5-三苯基氯化四氮唑(TTC)染色检测脑梗塞体积。结果TanⅡA治疗组脑梗塞体积较缺血再灌注组减小,高、低剂量组之间差异亦具有显著性(P<0.05);与缺血再灌注组比较,TanⅡA治疗组显著降低MPO活性、MDA含量,增加SOD、ATP含量,降低脑组织含水量,高、低剂量组之间差异亦具有显著性(P<0.05)。结论TanⅡA对缺血再灌注脑损伤具有保护作用,其机制可能与减轻缺血再灌注损伤炎症反应,减轻氧化性损伤和改善能量代谢有关。Objective To study the effect of Tanshinone Ⅱ A ( Tan Ⅱ A) on neutrophils infiltration and the contents of free radical and ATP in ischemic frontal and parietal cortex of cerebral ischemia reperfusion(I/R) rats. Methods In this experiment, rats were randomly divided into 4 groups, namely sham operated group, I/R group ,low dose Tan Ⅱ A treated group and high dose Tan Ⅱ A treated group. The focal middle cerebral artery occlusion(MCAO) model was made by suture - occluded method. Rats were pretreated with Tan Ⅱ A ig for 3d,respectively. After 90min MCAO following 24h of reperfusion, the MPO activity, the contents of SOD and MDA in ischemic frontal and parietal cortex were investigated. TTC staining was used to calculate the cerebral infarct volume. Results Compared with I/R group,cerebral infarction volume in low close Tan Ⅱ A treated group and high dose Tan Ⅱ A treated group decreased in a dose - dependent manner ( all P 〈 0.05 ). Compared with I/R group, low and high dose Tan Ⅱ A treated groups dose- dependently reduced MPO activity,the content of MDA and brain water content, increased the activity of SOD and the content of ATP. Conclusion Tan Ⅱ A may aueviate cerebral ischemia - reperfusion injury by decreasing the inflammation reaction , free radicals and the disbanlance of energy metabolism.
