摘 要:目的筛选超临界CO2流体萃取荜澄茄挥发性成分的最佳工艺,并研究萃取物的药效。方法采用正交实验设计,以萃取物得率为指标,考察萃取压力、萃取温度、解析压力及解析温度4个因素的影响;采用体外抗菌实验、抗致敏豚鼠回肠肌过敏性收缩反应实验、醋酸扭体法实验、小鼠自主活动次数影响试验研究萃取物的主要药效。结果最佳萃取工艺条件为萃取压力25MPa,萃取温度40℃,解析压力8MPa,解析温度50℃;萃取物具有显著的抑菌、抗过敏、镇痛、镇静等药理作用。结论超临界CO2流体萃取技术用于荜澄茄挥发性成分提取,方法简单可行,适用于工业化生产。荜澄茄超临界萃取物具有多方面的药理作用,研究开发价值较大。Objective To study the optimal technology of supercritical - CO2 fluid extraction ( SFE - CO2 ) for Litsea cubeba ( Lour. ) Pres. and its pharmacodynamic effect. Methods The yield of extract was used as index to evaluate the technologies based on orthogonal design , in which 4 factors considered were extraction temperature and pressure , separation temperature and pressure;the animal models of bacteriostasis, anti -anaphylaxis, analgesia, conscious -sedation were used to study the function of the extract. Results The optimum extraction conditions was extraction pressure 25MPa , temperature 40℃, separation pressure 8MPa, temperature 50% ; The extract had good pharmacodynamic action of bacteriostasis,anti -anaphylaxis, analgesia and conscious - sedation. Concision Supercritical fluids extraction method is feasible for the activities of Litsea cubeba ( Lour. ) Pres. , and useful for the large scale production. The extract has significant functions in many ways and is worthy to further development.
