高效液相色谱法测定复方元胡止痛胶囊中延胡索乙素的含量

摘　　要：目的延胡索乙素作为对照品，建立高效液相色谱（HPLC）法控制复方元胡止痛胶囊的质量。方法采用Alltima C18色谱柱（5μ0m，4、6mm×250mn），用甲醇-水-三乙胺（65：35：0、5）为流动相，检测波长280nm；柱温30℃；流速1、0ml／min。结果延胡索乙素含量在0、1029～1．0288μg范围内呈良好线性关系，得回归方程Y=4、8401X＋0、0895，r=0．9997，平均加样回收率及RSD分别为101、17％和1．02％。结论以延胡索乙素为对照品，采用HPLC法控制复方元胡止痛胶囊质量的方法简便快速、准确可靠、重复性好，专属性强。Objective To establish the method for determining the content of tetrahydropalmatine in complex Yuanhu Zhitong capsule by HPLC, Methods The separation was performed on Alltima C18 column （5 μm, 4.6mm × 250mm） with methanol - H2O - triethylamine （65： 35： 0.5） as mobile phase . The detection wavelength was at 280 nm, the column temperature was 30 ℃, and the mobile phase speed was 1.0ml per min. Results The calibration curve was linear in the range of 0. 1029 to 1. 0288 μg for the tetrahydropalmatine content and the linear equation was y = 4. 840 1x ＋ 0. 089 5 （ r = 0,999 7 ）. The average recovery rate was 101.17% and the relative standard deviation was 1.02%. Conclusion The method is simple, rapid, accurate and reproducible. It can be used for the determination of tetrahydropalmatine content complex Yuanhu Zhitong capsule.
