摘 要:目的建立测定人血浆中氢氯噻嗪浓度的LC-MS法。方法采集到的血浆样品加人对乙酰氨基酚为内标,经乙酸乙酯提取,进行LC-MS法测定。色谱柱为Kromasil C_(18)(150 mm×4.6 mm,5μm),流动相为甲醇-水(30:70,V/V),流速为0.8 mL·min^(-1),进样量为20μL,APCI选择负离子检测。结果本研究所建立测定氢氯噻嗪血浓度的LC-MS法,其线性范围为2.0~200μg·L^(-1),最低定量限为2.0μg·L^(-1),日内、日间RSD分别<8.6%、10.6%,均符合生物样品分析标准。结论该法灵敏度好、准确度高,可用于氢氯噻嗪的药动学研究及临床药物浓度检测。A/M To develop a LC-MS method for determination of hydrochlorothiazide (HCTZ) in human plasma. METHODS After adding intemal standard, the plasma samples were extracted and concentrated before being injected onto a reversed phase Kromasil C18( 150 mm × 4.6 mm, 5μm) column. The mobile phase was methanol-deionized water (30:70, V/ V) and the flow rate was 0.8 mL·min^-1; the injection vohmae was 20μL. Atmospheric pressure chemical ionization (APCI) source was applied and operated in negative ion mode. RESULTS The linear calibration curve was obtained in the concentration range of 2.0 - 200μg· L^-1 ( r = 0. 995 4). The limit of quantization was 2.0 μg· L^-1. The coefficient variations of intra-and inter-day were less than 8.6% and 10.6% respectively. CONCLUSION This method can be applied to determine hydrochlorothiazide in human plasma.
