摘  要:采用免疫胶体金定位和电子显微技术研究了离体条件下梨花柱S-RNase对自花、异花花粉管中钙调素(CaM)分布的影响.结果显示:(1)花粉管中CaM主要分布在质膜附近,在细胞壁上也有少量分布.(2)不亲和(自花)花柱S-RNase处理后花粉管远离质膜的位置发现CaM,处理24h后在花粉管中部出现CaM,且CaM位置向花粉管内部移动;而亲和(异花)S-RNase处理后花粉管的CaM分布没有明显变化.研究表明,CaM参与了自交不亲和反应过程.The variation of stylar S-RNase of pear on the calmodulin localization of compatible and incompatible pollen tube was studied in vitro by immuno-gold electron microscopy. The results showed that gold particles of CaM were found to be localized near by the plasma membrane. In addition, there were some CaM on cell wall. CaM was far from plasma membrane in incompatible pollen and it was found in the middle part of pollen tube after S-RNase deal 24 h. There was no evident variation of the calmodulin localization in compatible pollen tube with the same treatment. It showed that CaM was concerned with self-incompatibility, and offered evidence for the function of CaM in self-incompatibility.
