摘 要:目的探讨姜黄素诱导人红白血病细胞(K562)凋亡机制及其对细胞周期各时相变化。方法应用噻唑蓝(MTT)比色法和流式细胞术检测不同浓度的姜黄素对K562细胞增殖抑制率。细胞周期及凋亡率变化,并通过电子显微镜观察其亚细胞结构改变。结果姜黄素对K562有明显的增殖抑制作用,且呈剂量依赖性,流式细胞术结果显示,G1期细胞增多,S期细胞减少,G2/M期细胞相对增多,电子显微镜结果显示,细胞空化,线粒体肿胀,染色质趋边凝集。结论姜黄素对K562细胞有明显的抑制作用,阻止G1期细胞向S期细胞转化进程,诱导K562细胞凋亡。Objective Investigates the mechanism of curcumin-inducod K562 apoptosis and its affect on coil cycle phases. Methods Measured the survival rates,ceil cycle and apoptosis rates of K562 by treatment with various concentration of curcumin by the method of MTr and flow cytometry, and then observed the changes of its subceilular structure by metal microstructure microscope. Results Curcumin showed the significant effect on inhibiting the ceil proliferation of K562 in a dose-dependent manner. Flow cytometry showed the cells were increase in G1 stage and decrease in S stage, which were relative increase in G2/M stage. Electron microscopy examination indicated ceil cavitation, mitochondria swelling and chromatin edge agglutination. Conclusion Curcumin showed the significant effect on inhibiting the coil proliferation of K652, and hold back ceil course from G1 to S, and induced K562 apoptosis.
