摘  要:目的了解七叶皂苷钠(EscNa)对人脐静脉内皮细胞(HUVEC)活性及细胞周期分布的影响。方法在体外原代培养的HUVEC中加入不同浓度EscNa及肿瘤坏死因子(TNF-α),孵育24 h后,采用MTT和中性红法对HUVEC活性进行定量分析,流式细胞技术对细胞周期进行分析。结果MTT法和中性红法显示EscNa的半数抑制浓度(IC50)为(42.89±3.42)和(44.19±3.58) μg·mL^-1。10~80 μg·mL^-1浓度范围内的EscNa可不同程度地抑制HUVEC生长,呈浓度依赖性。细胞周期分析结果也显示,EscNa可阻抑G0/G1期细胞向S期的转变,抑制HUVEC增殖。结论七叶皂苷钠通过降低HUVEC活性,抑制细胞增殖,从而对HUVEC造成损伤。Objective To observe the effect of sodium aescinate (Esc-Na) on the activity of human umbilical vein endothelial cell(HUVEC) and the cell cycle. Methods Cultured HUVEC were treated with Esc-Na or tumor necrosis fator-α (TNF-α) at different concentrations for 24 hours. MTT method and the neutral red method were employed to detect the cellular activity. The cell cycle of HUVECs was measured by flow cytometry. Results The results showed that the half-maximal inhibiory concentrations(IC50) of Esc-Na were(42.89 ±3.42) for MTT and( 44.19 ±3.58) μg · mL^-1 for neutral red methool. At the concentration between 10 to 80 μg · mL^-1 , Esc-Na inhibited the growth of HUVEC in a concentration dependent manner. It was also showed that Esc-Na decreased the S-phase ceils and reduced G2 · M phase ceils to inhibit the proliferation of HUVEC. Conclusion Esc-Na exerted the cytotoxic effects to HUVEC by decreasing the cellular activity and inhibiting the cellular proliferation.
