摘 要:目的观察氧化苦参碱对单侧输尿管梗阻(UUO)大鼠模型转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)表达水平的影响。探讨氧化苦参碱对大鼠肾间质纤维化的干预作用,并阐明其可能的机制。方法将48只雄性SD大鼠随机分为假手术(SOR)组、UUO组、血管紧张素Ⅱ受体拮抗剂氯沙坦组(LOS组)和氧化苦参碱组(OM组)。检测血肌酐,肾组织行苏木精-伊红和Masson染色,免疫组化检测TGF-β1、α-SMA在肾组织中的表达,实时荧光定量PCR(RT-PCR)检测肾组织中TGF-β1和α-SMA的mRNA水平。结果LOS组和OM组大鼠的血肌酐与UUO组相比差异有统计学意义(P<0.05),但LOS组和OM组间无统计学意义(P>0.05);OM组和LOS组中各时间点TGF-β1、α-SMA的表达均分别低于UUO组(P<0.05),但高于SOR组(P<0.05),两治疗组间差异无统计学意义(P>0.05)。结论氧化苦参碱可从一定程度上减轻UUO大鼠肾间质纤维化程度,其作用机制可能为下调TGF-β1和α-SMA表达,从而减少细胞外基质的沉积,延缓肾间质纤维化。Objective To observe the effect of oxymatrine on renal tubulointerstitium in unilateral urteral obstruction(UUO)rat,and then exploer its possible mechanism.Methods 48 Sprague-Dawley male rats were randomly divided into sham-operated rats group(SOR group),unilateral ureteral obstruction model group(UUO group),losartan group(LOS group)and oxymatrine group(OM group).The levels of serum creatinine were examined at each time point.We observed the pathologic changes of the kidney and detected the expression levels of TGF-β1、α-SMA by immunohistochemistry and RT-PCR at each time point.Results The level of serum creatinine in UUO group was obviously higher than that in other groups at day 7,14(P〈0.05);The pathologic damage of the interstitium of UUO group was heavier than that of SOR group and treatment groups at the same time point.The expression levels of TGF-β1、α-SMA in OM and LOS groups were lower than that in UUO group at the same time point(P〈0.05).Conclusion oxymatrine can partly delay the progress of renal interstitial fibrosis in UUO rat model,its mechanism may be related to suppressing the expression of TGF-β1 and α-SMA,then reducing of renal interstitial fibrosis.
