摘  要:目的:检测大鼠胰腺B细胞中内源性大麻素受体1(CB1R)的定位,了解肥胖大鼠胰腺组织中CB1R表达的改变。方法:高脂饲料喂养雄性SD大鼠20周,制备肥胖大鼠模型(n=10),并设正常对照组(n=10)。取2组大鼠胰腺,免疫组织化学法检测CB1R蛋白表达,原位杂交法检测CB1RmRNA表达,间接双重免疫荧光染色法检测胰岛B细胞CB1R的定位。结果:CB1R主要表达于胰岛B细胞;且肥胖组大鼠胰腺组织CB1R蛋白及mRNA表达均高于对照组(P均<0.01)。结论:CB1R主要表达于胰岛的B细胞,且在肥胖时表达水平增加。Aim: To determine the existence of endocannabinoid receptor 1 ( CB1 R)in B cell and explore its expression changes in pancreas tissue of high-fat-diet-induced obese rats. Methods:A total of 20 male SD rats were randomly divided into 2 groups:one group was fed with high-fat-diet as obese rat model,the other group was fed with regular diet as normal control group. The pancreases of two groups were obtained after twenty weeks. The protein expression of CB1R was detected by using immunohistochemical method. The mRNA expression of CB1R was detected by using in situ hybridization method. Indirect double immunofluorescence was used to locate CB1R in the B cell of pancreatic islets. Results: Both immunohisto- chemistry and in situ hybridization methods revealed that CB1R were observed in the islets. And expression of CB1R in the islets of obese rats was higher than that of normal control group(P 〈0. 01 ). Conclusion: CB1R expresses in the pancreas islets and B cells are the chief expressing cells. CB1R expression in the islets of obese ones increases.
