甘遂提取物对肿瘤瘤株Hep、S_(180)的抑制作用观察

摘　　要：目的观察甘遂提取物作用于小鼠移植瘤细胞Hep、S180后的抑制作用。方法模型组、5-Fu组、甘遂提取物组(E)、5-Fu+E组,每组10只小鼠分别于右前肢腋皮下接种瘤中注射Tween80溶液、5-Fu、甘遂提取物、甘遂提取物+5-Fu。免疫组化检测Bcl-2表达。体外培养S180细胞,加入药物浓度为50μg/ml、10μg/ml、1μg/ml,培养48h,流式细胞仪测细胞凋亡率。结果模型组与各实验组移植癌和移植癌组织与药物注射引起肿瘤坏死的Bcl-2蛋白表达,在统计学上有显著差别(P<0.05)。加入药物浓度为50μg/ml、10μg/ml、1μg/ml,流式细胞仪测定凋亡率分别为33.9%、28.8%和23.3%。结论中药甘遂对Hep、S180有明显的抑制作用。Objective To observe the inhibitory effect of Euphorbia kansui on Hep and S180 cells tumor. Methods There were 10 of Hep and S180 in Euphorbia kansui group,10 in 5-Fu group, 10 in Euphorbia kansui and 5-Fu group, 10 in normal saline group. Expression of Bel-2 protein in those groups were measured with immunohistochemistry. Culture in vitro S180 cells and the inhibitory effects of Euphorbia kansui extract on S180 cells were determined the apoptosis rate of S180 cells by flow cytometry （FCM）. Results After being challenged with S180 and hep cells, the expression of Bcl-2 protein in the therapeutic group was lighter than that of the normal saline group （P〈0.05）.At a dose of 1.0μg/ml, 10.0μg/ml and 50.0μg/ml for 48h, the apoptosis rate was 23.3%,28.8% and 33.9% by FCM. Conclusion Euphorbia kansui may inhibit Hep and S180 efficiently.
