摘 要:目的研究糖尿病性脑病与海马区ICAM-1表达的关系,及银杏叶提取物(EGb)治疗的效果和机制。方法60只大鼠随机分为对照组、模型组和治疗组,各组再分为1个月和3个月两个时间点。以STZ制成糖尿病大鼠模型,治疗组应用EGb治疗,水迷宫试验观察大鼠行为学改变,免疫组织化学方法检测海马ICAM-1蛋白表达,RT-PCR检测ICAM-1 mRNA表达水平。结果(1)水迷宫试验发现模型1月组即已出现轻度学习、记忆功能障碍,3月组更加加重,治疗组则明显减轻。(2)对照组未见ICAM-1蛋白表达,模型组海马区呈阳性表达,治疗组表达减弱。(3)模型1月组海马区ICAM-1 mRNA表达升高,3月组更加明显,治疗组则显著减轻。结论海马区ICAM-1表达升高是糖尿病性脑病发病的重要因素,银杏叶提取物可能通过下调ICAM-1表达,而对糖尿病性脑病有保护作用。Objective To study the relation between diabetic encephalopathy and hippocampal ICAM-1 expression, and to investigate the therapeutic effect and mechanism of Ginkgo biloba extract on diabetic encephalo pathy. Methods Sixty rats were assigned into control group, model group and EGb-treatment group randomly, and every group was further divided into 1 month and 3-month subgroups. STZ was used to make the model of diabetes mellitus,and in EGb-treatment group EGb was abdominally injected every day. The behavioral abnor- malities were investigated by water maze test. The ICAM-1 expressions of mRNA and protein were evaluated by RT-PCR and immunohistochemistry method. Results (1) By maze water test, mild dysfunction of learning and memory was detected in model 1m group, and the dysfunction became worse in model 3m group. The matters were improved in EGb-treatment groups. (2) No expression of ICAM-1 was spotted in control groups. In the model groups, the expressions of ICAM-1 were positive in hippocampus,and the expression were weaker in EGb-treatment groups. (3) Compared with control group, the ICAM-1 mRNA level of hippocampus was increased in model lm group, and the level was obviously greater in model 3m group. In the EGb-treatment groups, the levels were dramatically reduced. Conclusion Diabetic encepha-lopathy may be closely related to hippocampal ICAM-1 expression. EGb has an protective effect on diabetic encepha-lopathy and the mechanism may be attributed to its effects by down-regu.lating the ICAM-1 expression.
