IGF-1和NF-^κB在家兔颈动脉粥样硬化斑块中的表达及其意义

摘　　要：目的:探讨IGF-1和NF-κB在家兔颈动脉粥样硬化斑块中的表达及分布特点。方法:先建立家兔颈动脉粥样硬化斑块性狭窄模型,之后应用免疫组织化学技术测定30例家兔颈动脉粥样硬化斑块中IGF-1和NF-κB的表达。结果:模型组家兔颈动脉粥样硬化斑块中IGF-1和NF-κB呈强阳性表达,且主要分布胞外基质中,偶见于泡沫细胞,胞核中NF-κB略有表达。而对照组家兔动脉壁中IGF-1和NF-κB仅在内中膜有微量表达。结论:检测颈动脉粥样硬化斑块中NF-κB及其靶基因IGF-1的表达变化,可以看出其激活与动脉粥样硬化病变的发生与发展密切相关,其在颈动脉粥样硬化斑块形成过程中发挥重要作用。Objective：To investigate the expression and distribution of insulin- like growth fatctor 1 （ IGF - 1 ） and nuclear factor - Kappa B （ NF -^κ B ） in 30 rabbits＇ atherosclerosis （AS） plaque of carotid artery cases. Methods： Firstly, a model of carotid atherosclerotic stenosis is established, and the protein level and area of IGF - 1 and NF - ^κB in AS plaque and normal rabbits＇ artery were studied by means of immunohistochemistry. Results：The expression of IGF - 1 and NF -^κB has a high level in AS plaque and mainly located in extracellular matrix, and also documented in foam ceils in AS plaque by chance. NF -^κB is also slightly expressing in nucleus. In contrast the expression of IGF - 1 was very low in normal arterial walls. Conclusion：The activation of NF -^κB and its target gene IGF - 1 is closely correlated in the forming and developing of AS ,and theexpressional changes of NF- ^κB ,IGF- 1 in the carotid AS plaque would play important role in carotid atherosclerotic plaque.
