兔胚泡肽合成片段及其与着床有关的生理功能

摘　　要：本研究测定了兔胚泡肽（RBP2）的氨基酸序列，它由54个氨基酸组成，其序列的第4位氨基酸至第34位氨基酸与兔子宫珠蛋白N端氨基酸序列完全相同。用计算机软件系统模拟分析，从54个氨基酸中选取了抗原性最高的一段氨基酸（aa20-aa34），然后人工合成了含有15个氨基酸的小肽片段（SPF）。研究表明：用溴脱氧尿嘧啶掺入淋巴细胞的方法测定了SPF对细胞增殖作用的影响，当浓度范围在80μg/ml至320μg／ml时，SPF的免疫抑制作用不明显。与天然RBP2比较，后者有明显的抑制作用。此结果说明，SPF的免疫抑制效应明显低于RBP2。实验又证明SPF（浓度范围在45～180μg／ml培液）对兔子宫内膜PGF2α的分泌呈剂量相关的抑制作用。此外，用[3H]亮氨酸掺入法，证明SPF（浓度180μg/ml培波）对[3H]亮氨酸掺入子宫内膜有明显促进作用，此促进作用被蛋白质抑制剂放线菌素（achdine）所阻断。The amino acid sequence of purified rahbit blastoeyst peptide2 (RBP2 ) was analyzed. It contains 54 amino acids and it's sequence from aa4 to aa34 is the same as the N-terminal sequence of uteroglobin- An active antigenic region consisting of l5 aa residues (aa2O-aa34) of the RBP2 was selected and designated as a synthetic peptide fragInent (SPF). The effect of SPF on lymphocyte proliferation was analyzed by immunocytochemical assay detecting BrdU incorporation in the cellular DNA. The results indicated that no obvious inhibitory effect of SPF on T cell proliferation at concentrations ranging from 80 to 32O μg/ml was observed, while RBP2 at the concentrations ranging from 20 to 16O μg/ml had a significant effect in a dose dependent manner, suggesting that the immuno-suppressive effect of SPF is less than that of RBP2. We have also observed that SPF at concentrations ranging from 45 to 180 μg/ml inhibited PGF2.secretion in a dose dependent manner. On the other hand, the 3H-leucine incorporation of rabbit endometrium was enhanced by SPF (180 μg/ml) and this action was blocked by additionof protein inhihitory agent, actidine.
