EFFECT OF STRESSFUL STIMULATIONS ON HYPOTHALAMIC VASOPRESSIN mRNA LEVEL OF RATS

摘  要:实验在给予慢性应激刺激的SpapeDawlay雄性大鼠中进行,用核酸斑点杂交技术观察大鼠下丘脑组织中血管升压汞(AVP)mRNA水平变化,用异羟基洋地黄毒甙(DIG)标记的26个碱基长寡聚核苷酸作为检测探针。结果观察到:在给予电击足底结合噪声的应激刺激之后,尾动脉收缩压逐渐升高,到刺激第5天,刺激组与对照组动物之间尾动脉收缩压差别有统计学显著意义;到刺激第9天,刺激组尾动脉收缩压达最高值;以后各天,血压持续维持在高水平。给予动物电击足底结合噪声的应激刺激4d以后,下丘脑组织中AVP-mRNA水平与对照组水平无显著差异;而给予刺激6d或15d之后,刺激组动物下丘脑组织中AVP-mRNA水平较对照组动物有显著升高(6d:P<0.005;15d:P<0.001)。实验中还观察到,实验组与对照组动物之间血浆渗透压无显著差异。上述结果提示:电刺激足底结合噪声的应激刺激在引起大鼠血压升高的同时,还能使下丘脑组织中AVP-mRNA水平升高;AVP-mRNA水平升高的机制有待进一步的研究。Experiments were cdried out in shessAn foot shock and noise stimulated male Spmpe Daway ratS. 3'-end digovigenin-labeled 26 mer oligonucliohde the was used to detect the vasopessin (AVP) InRN in hppthaldric hssues. Dot blotting tecboqUe was used in the assessment of AVP mRNA level. The resultS showed that the tall systolic arterial pressare of rats incaxd gradually ther sbessful shmulallons of foot shock in cornbination with noise. The difference in tall systolic blood pressme between control and shmulated rats was stahshcally significant after 5-day shmulation. By the ninth day, the systolic blood pressare of the sbessed rats reached the highest level and drintalned thereafter for sevend days. No significant difference was observed in the AVPInRN level between conbol and sbessed rats by the time when shIInilated for 4 days, but it significanily increases when stimulated for 6 day or longer (by the 6th day: P < 0.005;by the l5th day: P < 0.001). No difference was foed in plasrna osmolalihes between conrml and sbessed AnInals. These resultS indicte'that the sbessfor foot shocks in coInbination with noise stimulations may cause an increase in AVPrnRN level in the hpothalams, which was accompanied by an incax in systolic arterial presare. Themecbosm of incare in AVP-mRNA level upon sbess shmulation edns to be elucidaed.
