摘  要:目的:观察雌激素对雌性大鼠颏舌肌细胞肌质网(sarcoplasmic reticulum,SR)Ca2+-ATPase活性及其基因表达的影响,探索其影响颏舌肌功能的分子生物学机制。方法:将30只成年雌性SD大鼠随机分为对照组(control)、去卵巢组(ovariectomy,OVX)、去卵巢回补激素组(ovariectomy with 17β-estradiol,OVX+E2)。术后6周,处死动物,提取颏舌肌SR。采用定磷法检测颏舌肌SR Ca2+-ATPase活性,荧光定量RT-PCR测定SR Ca2+-ATP酶mRNA表达的变化。结果:OVX组血清雌激素水平及子宫湿重较对照组明显降低(雌二醇P<0.01;子宫湿重P<0.01),而OVX+E2组与对照组相比差别无统计学意义(P>0.05)。OVX组颏舌肌SR Ca2+-ATP酶活性较对照组明显降低(P<0.01);OVX+E2组较OVX组有明显提高(P<0.01)、与对照组相比差别无统计学意义(P>0.05)。OVX组颏舌肌SR Ca2+-ATPase mRNA表达较对照组明显降低(P<0.01);OVX+E2组较OVX组有明显提高(P<0.01)、与正常组相比差别无统计学意义(P>0.05)。结论:成年雌性大鼠雌激素水平的变化可引起颏舌肌SR Ca2+-ATP酶活性及其mRNA表达的变化,雌激素可能通过该途径改变颏舌肌功能.Objective:To investigate the effects of estrogen on sarcoplasmic reticulum (SR) Ca^2+ -ATPase activity and gene expression in female rats genioglossal muscle (GG) , and explore the possible molecular biology mechanism. Methods:30 male rats were randomly divided into 3 groups:Control group, ovariectomy(OVX) group, ovariectomized animals receiving estradiol replacement ( OVX + E2 ) group. The rats were sacrificed 6 weeks after surgery, and the genioglossus was taken out for analysis. Ca^2+ -ATPase activity was measured by detecting inorganic phosphorus ion, and SR Ca^2+ -ATPase (SERCA) mRNA level was observed using fluorescence quantitative reverse transcription-polymerase chain reaction (FQ RT-PCR) method. Results:Compared to the control group, SR Ca^2+- ATPase activity of OVX group remarkably reduced( P 〈0.01 ), and SERCA mRNA level was also significantly lowered relevandy(P 〈0.01 ) ; while in OVX + E2 group, there were no significant changes in SR Ca^2+ -ATPase activity and SERCA mRNA level( P 〉 0.05 ). Conclusion: Serum level of estrogen of adult female rats may influence the function of genioglossal muscle by influencing SR Ca^2 + -ATPase activity and the SERCA mRNA expression.
