角质细胞生长因子对无排卵功血子宫内膜上皮细胞PCNA、ER和PR表达的影响

摘　　要：目的观察角质细胞生长因子(keratinocyte growth factor,KGF)对离体培养的无排卵型功血子宫内膜上皮细胞EEC中增殖细胞核抗原PCNA、雌激素受体ER和孕激素受体PR表达的影响。方法体外培养无排卵型功血子宫内膜上皮细胞,加入不同浓度的KGF作用后,免疫组化法检测PCNA、ER和PR的表达变化,RT-PCR检测ER和PR mRNA表达的变化。结果免疫组化染色结果,PCNA蛋白表达位于细胞核,ER和PR蛋白表达主要位于细胞核内,少数可见细胞质着色;50ng/ml KGF作用后,正常组和功血组PCNA表达分别较作用前的对照组显著增高,ER和PR表达分别较作用前的对照组显著增高(P<0.05);PCNA和ER表达的增加幅度在正常组和功血组之间无显著差异(P>0.05);而功血组PR表达的增加幅度显著高于正常组(P<0.05);50ng/ml KGF作用后,RT-PCR方法检测正常组和功血组ER和PR的mRNA表达,分别较作用前的对照组显著增高(P<0.05)。结论KGF对体外培养的无排卵型功血和正常的子宫内膜上皮细胞均有增加PCNA、ER和PR表达的作用,但增加程度有差异,提示两组的子宫内膜上皮细胞生长均受KGF的影响,但无排卵功血可能存在与正常的子宫内膜上皮细胞不同的生长机制。Objective To observe the effect of keratinocyte growth factor （KGF） on PCNA, ER and PR expressions of cultured endometrial epithelial cells （EECs） from the patients with anovulatory dysfunctional uterine bleeding （ADUB）. Methods EECs from the patients with ADUB were separated and cultured with KGF of different concentration, with the normal EECs as control. Proliferative cell nuclear antigen （PCNA）, estrogen receptor （ER）, and progesterone receptor （PR） were detected by immunocytochemistry. The mRNA expressions of ER and PR were determined by RT-PCR. Results PCNA was only expressed within nucleus. ER and PR proteins were mainly expressed within nucleus, and some also expressed in plasma. KGF （50 ng/ ml） increased PCNA, ER and PR expressions in both ADUB and normal EECs （P 〈 0.05 ）. KGF （50 ng/ml） increased the mRNA expressions of ER and PR in both ADUB and normal EECs （ P 〈 0.05）. The increment of PCNA and ER expressions in ADUB group was the same as that in normal group （ P 〉 0.05 ） , but the increment of PR expressions in ADUB group was more than that in normal group （ P 〈 0.05）. Conclusion KGF can increase PCNA, ER and PR expressions in cultured ADUB and normal EECs, but the increment in the two groups is significantly different, indicating that EECs from ADUB endometrium and normal endometrium react to the stimulation of KGF in different ways.
