摘  要:目的:进行大鼠骨髓间充质干细胞(MSCs)与成纤维细胞(FBs)细胞的蛋白质组学研究,寻找出重要的差异蛋白质。方法:采用二维电泳的方法(2DE)分别分离两种细胞的蛋白质,用PDQuest软件分析蛋白表达差异,并采用质谱(MALDI-TOF-MS)进行鉴定。结果:发现了MSCs和FBs之间存在差异表达的15种蛋白质,其中4种蛋白质差异显著且与细胞功能密切相关,即SM22-α、鸟嘌呤脱氨基酶(Guanine deaminase,GD)、膜联蛋白(Annexin A5)、过氧化物岐化酶-2(superoxide dismutase2,SOD-2)。MSCs的SM22-α和Annexin A5的表达分别比FBs低26倍和8倍,而MSCs的GD和SOD-2表达分别比FBs高16倍和21倍。结论:与大鼠FBs相比,MSCs存在更多防止细胞损伤相关的蛋白质和更少的与细胞衰老过程密切相关的蛋白质,具有高分化潜能,更适合于作为组织工程种子细胞。Objective To analysis and to compare the proteomic differences between mesenchymal stem cells(MSCs) and fibroblasts (FBs), and to discover the key different proteins. Methods Separate the protein of cells by 2 dimension electrophoresis (2DE), analysis the differences of protein expression with PDQues, and identify with MALDI-TOF-MS. Results Discover fifteen different types of proteins, four of which associate with different functions of MSCs and FBs. Those are SM22-alpha, Guanine deaminase(GD), Annexin A5 and superoxide dismutase 2 (SOD-2). the expression of SM22-αand Annexin A5 in MSCs is separately 26 folds and 8 folds lower than in FBs, but MSCs express GD and SOD2 16 folds and 21 folds higher than FBs. Conclusion Compared with FBs, MSCs express more kinds of proteins that can prevent the damage of themselves, while have fewer proteins associated with aging in expression. Plus pluripotent differentiation potential, they are better seed cells for tissue engineer.
