摘  要:目的探讨共培养体系中体细胞对胚胎早期发育的影响及比较2种常用共培养体系的优劣。方法实验分3组:同种不同类型体细胞,受精卵分别在含颗粒细胞和成纤维细胞的M199培养液中培养;异种同类型体细胞,受精卵分别在含牛颗粒细胞和小鼠颗粒细胞中M199培养液中培养;2种共培养体系比较,受精卵分别在"M199+颗粒细胞"和"B2+Vero细胞"的共培养体系中培养。结果颗粒细胞组较成纤维细胞组囊胚率高(P<0.05);牛颗粒细胞组和小鼠颗粒细胞组在卵裂率和囊胚率方面差别无统计学意义;"M199+颗粒组"在卵裂率和囊胚率方面与"B2+Vero组"差别无统计学意义,但囊胚细胞数较少(P<0.05)。结论在一定的条件下,共培养体系中体细胞对胚胎早期发育有一定影响;"B2+Vero细胞"的共培养体系优于"M199+颗粒细胞"的共培养体系。Objective To study the effect of somatic cell co-culture system on the early bovine embryo development and comparison of two different co-culture systems. Methods Experiment l:the somatic cells were of the same breed but different types (granule cell group and fibroblast group); experiment 2. the somatic cells were of the same type but different breeds(bovine granule cell group and mouse granule cell group) ; experiment 3: compare two co-culture system(M199+granule cell group and B2+Vero ceils group). Results In experiment 1, granule cell group was similar to fibroblast group in cleavage rate but higher in blastocyst rate~ in experiment 2,there was no difference between two groups in cleavage rate and blastocyst rate; in experiment 3, there was no difference between two groups in cleavage rate and blastocyst rate, but total cell number of blastocysts in B2+Vero cells group was higher than that of M199 +granule cell group. Conclusion Somatic cell co-culture system may have an effect on the early embryo development some conditions, and the co-culture in "B2+Vero cells" is better than in "M199 +granule cells" for bovine early embryo development.
