摘 要:应用PCR-RFLP技术对来自四川省8个地方的枯死松树样品中的伞滑刃属线虫(Bursaphelenchus)群体进行了分子鉴定。结果显示来自邻水、雅安的枯死松树样本中含有松材线虫(B.xylophilus),而来自大竹和遂宁的枯死松树样本中含有拟松材线虫(B.mucronatus)。其他伞滑刃属线虫包括莱鲁尔夫伞滑刃线虫(B.rainulf)、霍夫曼伞滑刃线虫(B.hofumanni)、泰国伞滑刃线虫(B.thailandae)、变异伞滑刃线虫(B.aberrans)。本研究中选用的4种限制性内切酶(DraⅠ、SalⅠ、HhaⅠ、AluⅠ)除SalⅠ外均可对所鉴定的这6种伞滑刃属线虫产生特异性的酶切位点,从而可均有效地鉴别出松材线虫、拟松材线虫和其他4种伞滑刃属线虫,其结果可为今后这几种伞滑刃属线虫的分子鉴定提供一定的参照。A ploymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis was used to differentiate eight kinds of Bursaphelenchus. The samples were gathered from 8 counties in Sichuan province. The results showed that the samples from LS and YA were validated as B. xylophilus; the samples from DZ and LS were validated as B. mucronatus; and the others were validated as B. rainulf, B. hofurnanni, B. thailandae. Four kinds of restriction enzymes (Dra Ⅰ .Sal Ⅰ .Hha Ⅰ .Alu Ⅰ) produced specific restriction sites, so that the enzymes selected in trial can differentiate B. xylophilus and t3. mucronatus, from the others except Sal Ⅰ. The result will offer reference to identify Bursaphelenchus by molecular means.
