酚染色法检验小麦品种纯度的可靠性分析

摘  要:用1%邻苯二酚溶液处理20个小麦品种(系)进行纯度检验,并与田间纯度检验进行比较,发现过高地估计了品种纯度;而用0.125%邻苯二酚溶液处理这20个品种,又过低地估计了品种田间纯度。对另外4个品种用0.125%邻苯二酚处理,染成浅色和黑色种子进行穗发芽抗性鉴定,发现与PPO活性密切相关的穗发芽抗性存在较大的变异。对18个小麦杂交组合的F2种子用0.125%邻苯二酚处理,分别将浅色和黑色F2种子种植并测定株高,结果表明:PPO活性与株高并无相关性。实验证明酚染色法检验小麦品种纯度尚缺乏准确性,需进一步完善。Twenty wheat cuhivars or lines were treated with 1% catechol solution to test their cultivar purities, the results were higher than those tested in field. When the twenty wheat cuhivars or lines were treated with 0. 125% catechol solution, their cuhivar purities were lower than those tested in field. The variance of pre-harvest sprouting (PHS) resistance, closely related to polyphenol oxidase (PPO), was found, when another four wheat cuhivars were treated with 0. 125% catechol solution and identified PHS resistance of light color and dark black seeds. F2 seeds derived ,from eighteen wheat cross combinations were treated with 0. 125% catechol solution and plant heights of light color and dark black seeds were determined, the result showed that PPO activity was not related to plant height. Using PPO pigmentation method in National Rules for Seed Testing (GB/T 3543.5-1995 ) ,the tested cultivar purity was not exact. The method needs consummated.
