摘  要:目的:观察腺病毒介导人骨形成蛋白7基因(Human bone morphogenetic protein-7,hBMP-7)转染骨髓间充质干细胞(Bone mesenchymal stem cell,BMSC)后移植兔退变椎间盘,对退变椎间盘髓核组织中蛋白多糖含量的影响。方法:20只新西兰大白兔手术建立椎间盘L3/4、L4/5、L5/6退变模型,随机分为两组,每组10只:实验组L3/4、L4/5、L5/6椎间盘内注射腺病毒介导hBMP-7基因转染的BMSC;对照组L3/4、L4/5、L5/6椎间盘注射磷酸盐缓冲液(PBS)。术后连续行磁共振观测椎间盘变化;12周后取材,应用RT-PCR、免疫组化检测hBMP-7的表达情况,分光光度法测量蛋白多糖含量。结果:与对照组相比,实验组的磁共振T2信号强度以及蛋白多糖含量均明显增高(P<0.05)。hBMP-7的mRNA和蛋白水平在实验组中的表达水平也最高。结论:腺病毒介导hBMP-7转染BMSC后,移植退变椎间盘,可使髓核组织中蛋白多糖的含量增加。Objective: To study the effect of bone mesenchymal stem cell (BMSC) transferred by hBMP-7 gene on proteoglycan of degenerative intervertebral disc of rabbits. Methods: Models of degenerative intervertebral disc were established on 20 rabbits and the animals were randomly divided into two groups. For study groups, BMSC transferred by adenovirus vector mediated hBMP-7 gene was injected into degenerative intervertebral disc; for control groups, PBS was injected into the same intervertebral disc. The changes of the intervertebral disc was showed by MRI, and the proteoglycan was quantitated by spectrophotometer to observe the repair effect. The hBMP-7 expression was detected by immunohistochemistry and RT-PCR in vivo. Results: The MRI T2 value and the content of proteoglycan of study groups had significant difference comparing with that of control grup (P〈0.05), as well the expression level of hBMP-7 mRNA and protein. Conclusions: BMSC transferred by adenovirus vector mediated hBMP-7 gene are helpful for improving the expression of proteoglycan in nucleus pulposus after injecting into degenerative intervertebral disc of rabbits.
