脑缺血再灌流后大鼠大脑皮质微血管密度的体视学

摘  要:为了研究脑缺血再灌流后大鼠大脑皮质微血管的变化,采用24只SD大鼠,分为对照组与缺血再灌流后6h、12 h和24 h组。以碱性磷酸酶法显示微血管,体视学方法测算大鼠海马、齿状回及顶叶皮质微血管密度的变化。结果表明:脑缺血再灌流后大鼠大脑海马、齿状回及顶叶皮质微血管的长度密度(Lv)、表面积密度(Sv)、体积密度(Vv)发生显著变化,尤其在再灌流后6、12 h。与对照组相比,Sv在6、12、24 h持续下降(P<0.01),Lv在6 h极显著下降(P<0.01),12 h极显著上升(P<0.01),而Vv在6 h极显著上升(P<0.01),12 h显著下降(P<0.05),24 h组Lv、Vv同对照组相比无显著性变化(P>0.05)。与6 h相比,12 h组齿状回及顶叶皮质区微血管Vv具极显著性差异(P<0.01),24 h组大脑皮质微血管Vv具极显著性差异(P<0.01)。各组中同区左右两侧比较微血管密度均不存在显著性差异(P>0.05)。各组中顶叶皮质区微血管密度高于海马及齿状回(P<0.01)。In order to investigate the changes of cerebral cortex microvessel after cerebral ischemia -reperfusion ,24 SD rats were randomly divided into control group, ischemia -reperfusion 6h, 12h and 24h group. The microvessel of hippocampus ,dentate gyrus and parietal cortex were observed by Alkaline phosphatase (AKP) method and micro- vascular density changes were measured by stereology. The results showed that the microvascular length density (Lv), surface density (Sv) and volume density (Vv) varied significantly after ischemia -reperfusion, especially at the period during reperfusion 6h and 12h. Compared with control group, Sv decreased (P 〈0.01 ) continually from 6h to 24h,Lv decreased significantly (P 〈0.01 ) at 6h and increased significantly (P 〈0.01 ) at 12h,while Vv in- creased significantly (P 〈0.01 ) in 6h and decreased (P 〈0.05) in 12h,and there were no significant differences ( P 〉 0.05 ) between control group and 24h group in Lv and Vv. Compared with 6h group, there was significant differences ( P 〈0.01 ) at 12h in Vv of dentate gyrus and parietal cortex,and there was significant differences ,( P 〈 0.01 ) at 24h in Vv of cerebral cortex. The microvascular density between the left and right side of the same part in each group showed no significant difference ( P 〉 0.05 ). The microvascular density of parietal cortex was the highest (P 〈0.01) in each group compared with hippocampus and dentate gyrus.
