摘  要:目的研究雌激素受体卢亚型(estrogen receptor β,ERβ)选择性激动剂广藿香醇(patchouli alcohol,PA)对β-淀粉样蛋白(β-amyloidpe ptide,Aβ)片段神经毒性的体外抑制作用。方法Aβ25-35位氨基酸片段(Aβ25-35)作用于人神经母细胞瘤细胞(SH-SY5Y)之前不同浓度PA预防给药,荧光探针法检测细胞内活性氧(reactive oxygen species,ROS)水平和细胞内钙离子浓度([Ca^2+];),流式细胞术检测细胞凋亡率。结果PA(0.05、0.5、5μg·ml^-1)能够抑制Aβ25-35引起的细胞内ROS水平和[Ca^2+];升高,使细胞凋亡减少。结论PA对Aβ25-35的神经毒性具有抑制作用。Aim To observe the activities of selective estrogen receptor (ER)β-agonist, patchouli alcohol (PA), on neurotoxicity of β-amyloid peptide fragment 25 -35 (Aβ 25-35 ). Methods SH-SYSY cells were pretreated with indicated concentrations of PA prior to β -amyloid peptide fragment 25 -35 (Aβ 25-35 ) exposure. Then the intracellular reactive oxygen species (ROS) level and intracellular calcium concentration ([ Ca^2+ ]i) were detected using the fluorescence probe, and the apoptosis rate of the cells was detected by flow cytometry. Results Pretreatment with PA (0.05,0.5,5μg·ml^-1) markedly attenuated Aβ 25-35-induced increases of the ROS level and [ Ca^2+ ]i,resulting in the decrease of the apoptosis rate. Conclusion PA can protect against Aβ 25 -35-induced toxicity.
