摘 要:目的建立六味地黄口服液中多糖的含量测定方法,比较微滤、超滤和醇沉法对多糖含量的影响。方法采用苯酚-硫酸显色法测定六味地黄口服液中多糖的含量。结果样品经苯酚-硫酸显色后于490nm处测定吸收度,在此波长处溶液的吸收度与葡萄糖含量呈良好线性关系。线性范围:12.0—72.0μg·ml^-1,r=0.9998,加样回收率为99.04%,RSD为1.16%(n:6);3种工艺六味地黄口服液多糖含量以葡萄糖计分别为8.22、7.04、7.24mg·ml^-1。结论苯酚-硫酸显色法操作简便,准确可靠,可作为六味地黄口服液质量控制方法之一,经孔径为0.18μm微滤膜处理后的六味地黄口服液中多糖含量最高。Aim To establish a method for the determination of polysaccharides in LOL and to study the determinations of LOL using microfiltration method, ultrafiltration method and ethanol-sedimentary method. To provide some reference for quality control of LOL. Methods Using Phenol-sulfuric Method to determine the contents of polysaccharides in LOL. Results The absorptions of processed samples were tested at the wavelength of 490nm. The linear range of Glucose was 12.0 -72.0μg·ml^-1, r =0. 999 8;the average recovery was 99.04%, RSD = 1.16% ( n =6). The contents of polysaccharides of LOL as produced by the three different proccesses were 8.22,7.04 and 7.24mg·ml^ - 1. glucose respectively. Conclusion Phenol-sulfuric acid Method is reliable, and can be used of as a routine method. Contents of polysaccharides in LOL filtrated through microfiltration membrane with a pore size of 0. 18μm were relatively high.
