摘  要:目的探讨运动干预对大鼠骨骼肌磷脂酰肌醇3-激酶(PI3K)mRNA表达、蛋白总量(t-PI3K)及磷酸化PI3K(p-PI3K)的影响。方法SD大鼠随机分为对照组和运动组。运动组分别进行1h/d和1.5h/d的游泳运动7周,最后一次运动结束恢复24h和48h取材,分为1h/d运动24h取材组、1h/d运动48h取材组、1.5h/d运动24h取材组、1.5h/d运动48h取材组。测定血液葡萄糖和胰岛素浓度;Westernblot法检测骨骼肌t-PI3K蛋白表达、p-PI3K磷酸化水平;RT-PCR法分析PI3KmRNA表达。结果与对照组比较,运动组胰岛素浓度降低;各运动组p-PI3K升高;1h/d运动24h取材组和1.5h/d运动24h与48h取材组t-PI3K增高;PI3KmRNA表达变化发生在1h和1.5h运动24h取材组。结论运动干预提高大鼠骨骼肌对胰岛素的敏感性,改善PI3K磷酸化、蛋白和基因表达。Objective To investigate the effects of exercise on PBK phosphorylation,protein and gene expression. Methods Male rots were randomly divided into control and trained groups. The trained rots exercised for 1 or 1.5 h daily for 7 weeks and had a fragment of their excised gastroonemius muscle,24 or 48 h after the last training session. The rats were divided into 1h/d exercise group of recovery 24 h,1 h/ d exercise group of recovery 48 h, 1.5 h/d exercise group of recovery 24 h and 1.5 h/d exercise group of recovery 48 h. The serum levels of glucose and insulin were measured. The changes in the expressions of PI3K and p-PI3K were determined by Western blotting. The expression of PI3K mRNA was determined by RT-PCR. Results Glucose tolerance test found that blood insulin concentration was decreased with exercise training. Exercise led to a marked increase in p-PI3K of trained groups as compared with controls, and the protein expression of PI3K significantly increased 24 h after exercise in rats that exercised for 1 h per day and 24 and 48 h after the exercise in those that exercised for 1.5 h per day. PI3K mRNA was increased by exercise 1 h/d and 1.5 h/d, 24 h after the last training session. Conclusion Exercise could increase muscle responsiveness to insulin, improving the total PI3K and p-PI3K and mRNA expression.
